HPLC-ELSD同时测定猴头菌丝固体培养物及胃乐宁片中麦角甾醇和β-谷甾醇的含量

目的:建立HPLC-ELSD同时测定猴头菌丝固体培养物及胃乐宁片中麦角甾醇和β-谷甾醇含量方法。方法:采用Kromasil C18色谱柱(4.6 mm×250 mm,5 μm),以甲醇为流动相,流速1 mL·min-1,柱温35℃,检测器为ELSD,漂移管温度70℃,载气压力275.8 kPa。结果:麦角甾醇和β-谷甾醇峰面积的对数与浓度的对数呈良好的线性关系(r>0.999 2),猴头菌丝固体培养物中麦角甾醇和β-谷甾醇平均回收率分别为92.6%和94.1%;胃乐宁片中麦角甾醇和β-谷甾醇平均加样回收率分别为103.0%和94.6%。10批猴头菌丝固体培养物中麦角甾醇和β-谷甾醇的含量分别为0.081~0.178和0.101~0.128 mg·g-1;10批胃乐宁片中麦角甾醇和β-谷甾醇的含量分别为0~0.252和0~0.380 mg·g-1结论:该方法可用于猴头菌丝固体培养物及胃乐宁片质量控制。

Objective: To establish an HPLC-ELSD method for determining the ergosterol and β-sitosterol in solid cultures of Hericium mycelium and Weilening tablets.Methods: HPLC analysis was performed on a Kromasil C18 column(4.6 mm×250 mm, 5 μm)with methanol as the mobile phase.Flow rate was set at 1 mL·min-1 and column temperature was 35℃.The analysis was done by ELSD with drift tube temperature at 70℃ and gas pressure at 275.8 kPa.Results: The logarithm of peak area of ergosterol and β-sitosterol were linearly correlatedx to the logarithm of injected mass within an acceptable range(r>0.999 2).The average recovery of ergosterol and β-sitosterol in solid cultures of Hericium mycelium were 92.6% and 94.1%,respectively.The average recovery of ergosterol and β-sitosterol in Weilening tablets were 103.0% and 94.6%,respectively.The contents of ergosterol and β-sitosterol in 10 samples in solid cultures of Hericium mycelium were 0.081-0.178 mg·g-1 and 0.101-0.128 mg·g-1; the contents of ergosterol and β-sitosterol in 10 samples of Weilening tablets were 0-0.252 mg·g-1and 0-0.380 mg·g-1.Conclusion: The established method can be applied to the quality control of solid cultures of Hericium mycelium and Weilening tablets.
