蒙古族药塔日努及其不同炮制品HPLC指纹图谱研究

目的:建立蒙古族药材塔日努及其牛奶制、酒制及诃子汤制品指纹图谱,为塔日努不同炮制品质量控制及炮制原理研究提供依据。方法:采用HPLC,Hypersil C18色谱柱(4.6 mm×250 mm,5 μm),流动相为乙腈-0.4%磷酸水,梯度洗脱,柱温40℃,检测波长254 nm,对塔日努及其3种炮制品进行指纹图谱研究,采用指纹图谱相似度软件进行分析。结果:建立了塔日努及其3种炮制品指纹图谱,塔日努经炮制后,其成分发生了变化,塔日努生品及牛奶、诃子汤制品的指纹图谱有明显区别,而生品塔日努及酒制塔日努指纹图谱较为相似。奶制品中5号锋消失,诃子汤制品中新增7号和8号锋、新增的7号峰峰面积较大。结论:该法精密度、重复性、稳定性良好,符合指纹图谱技术要求,可作为塔日努及其炮制品的质量控制方法。

Objective:To provide a basis for quality control and processing principle research of Tarinu and its different processed products by establishing HPLC fingerprints.Methods:HPLC was performed on a Hypersil C18 column (4.6 mm×250 mm,5 μm)with mobile phase of acetonitrile-0. 4% phosphoric acid in gradient elution at a flow rate of 1.0 mL·min-1.The column temperature was 40℃ and the detection wavelength was 254 nm. The fingerprints of Tarinu and its 3 kinds of processed products were analyzed by fingerprint similarity software.Results:There were significant differences in fingerprints of raw products and milk and Terminalia chebula Retz soup processing products. But fingerprints of raw products and alcohol processing products were relatively similar. Compared with the fingerprint of Tarinu,peak 7 and peak 8 were found as new components in Terminalia chebula Retz soup processing product and the peak area of peak 7 was relatively larger. On the other hand,peak 5 disappeared in milk processing product.Conclusion:The method is stable and accurate with good reproducibility and can be used in quality control of Tarinu and its processed products.
