目的:建立高效液相色谱法同时测定民族药材青阳参中对羟基苯甲酸、香草酸、对羟基苯乙酮、2,5-二羟基苯乙酮、白首乌二苯酮和2,4-二羟基苯乙酮的含量。方法:采用HSS T3 C18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈-0.05%甲酸水溶液(20:80)为流动相,流速1 mL·min-1,检测波长为260 nm,柱温30℃,进样量10 μL。结果:对羟基苯甲酸、香草酸、对羟基苯乙酮、2,5-二羟基苯乙酮、白首乌二苯酮和2,4-二羟基苯乙酮质量浓度分别在0.082~4.084 μg·mL-1(r=0.999 6)、0.080~4.024 μg·mL-1(r=0.999 8)、0.158~7.888 μg·mL-1(r=0.999 7)、0.078~3.900 μg·mL-1(r=0.999 7)、0.308~15.41 μg·mL-1(r=0.999 6)和0.077~3.872 μg·mL-1(r=0.999 8)范围内线性关系良好,平均加样回收率、精密度、重现性和稳定性均符合有关规定。样品中6个酚类成分的含量分别为对羟基苯甲酸0.019~0.133 mg·g-1,香草酸0.021~0.140 mg·g-1,对羟基苯乙酮0.068~0.300 mg·g-1,2,5-二羟基苯乙酮0.029~0.383 mg·g-1,白首乌二苯酮0.220~1.507mg·g-1,2,4-二羟基苯乙酮0.034~0.369 mg·g-1。结论:本法可用于青阳参中6个酚类成分的含量测定。
Objective: To establish an HPLC method for the simultaneous determination of 6 phenols, including p-hydroxybenzoic acid, vanillic acid, p-hydroxyacetophenone, 2, 4-dihydroxyacetophenone, baishouwubenzophenone and 2, 5-dihydroxyacetophenone in ethnic medicine Radix Cynanchi Otophylli.Methods: HPLC analysis was performed on a HSS T3 C18(250 mm×4.6 mm, 5 μm)column. The mobile phase was acetonitrile-0.05% formic acid solution(20:80)at the flow rate of 1 mL·min-1 and the detection wavelength was 260 nm. The column temperature was 30℃ and the injection volume was 10 μL.Results: The standard curves revealed a good linear relationship in the ranges of 0.082-4.084 μg·mL-1(r=0.999 6)for p-hydroxybenzoic acid,0.080-4.024 μg·mL-1(r=0.999 8)for vanillic acid,0.158-7.888 μg·mL-1(r=0.999 7) for p-hydroxyacetophenone, 0.078-3.900 μg·mL-1(r=0.999 7)for 2, 5-dihydroxyacetophenone, 0.308-15.41 μg·mL-1(r=0.999 6)for baishouwubenzophenone and 0.077-3.872 μg·mL-1(r=0.999 8)for 2, 4-dihydroxyacetophenone,respectively. The contents of 6 phenols were 0.019-0.133 mg·g-1 for p-hydroxybenzoic acid, 0.021-0.140 mg·g-1 for vanillic acid,0.068-0.300 mg·g-1 for p-hydroxyacetophenone,0.029-0.383 mg· g-1 for 2, 5-dihydroxyacetophenone, 0.220-1.507 mg·g-1 for baishouwubenzophenone and 0.034-0.369 mg·g-1 for 2, 4-dihydroxyacetophenone,respectively.Conclusion: The developed method is suitable for the quantitative analysis of six phenols in Radix Cynanchi Otophylli.
