HPLC-ELSD法测定不同产地麦冬中4种代表性成分的含量

目的:建立测定麦冬类药材中具有代表性的2个黄酮和2个皂苷成分的HPLC-ELSD方法,比较研究不同产地麦冬中指标成分的差异。方法:采用HPLC-ELSD法,使用Kromasil 100-5 C18(4.6 mm×250 mm,5 μm)色谱柱,流动相为乙腈(A)-水(B),梯度洗脱(0~60 min,35% A→65% A),流速1.0mL·min-1,柱温30℃,蒸发光散射检测,漂移管温度100℃,气体流速3.0 L·min-1,进样量20 μL。结果:在不同来源麦冬中的麦冬皂苷D、麦冬皂苷D’、麦冬甲基黄烷酮A、麦冬甲基黄烷酮B 4种成分色谱峰均达到基线分离,各成分均有较宽的线性范围和良好的线性关系(r>0.999),加样回收率在97.45%~103.4%。不同产地麦冬中4种有效组分的含量差异极显著(P<0.01),同一产地麦冬中4种有效组分含量存在一定差异,但多不显著。结论:建立的麦冬中代表性成分含量测定方法可用于麦冬药材质量评价和道地性分析。

Objective: To establish an HPLC method to measure the contents of 2 representative flavone constituents and 2 representative saponin constituents in Maidong type medicinal materials,and to study the difference among the Maidong medicinal materials from different producing areas.Methods: HPLC-ELSD was adopted in the determination.The analysis was performed on a Kromasil 100-5 C18 column (4.6 mm×250 mm,5 μm).The mobile phase was acetonitrile (A)-water(B) with gradient elution mode (0-60 min, 35%A→65%A) at the flow rate of 1.0 mL·min-1.The column temperature was 30℃,the detector was ELSD, the drift tube temperature was 100℃,the gas flow rate was 3.0 L·min-1,and the sample size was 20 μL.Results: The baseline of 4 constituents,including ophiopogonin D,ophiopogonin D',methylophiopogonanone A,and methylophiopogonanone B,in Maidong from different producing areas was well separated with wide linearity ranges and good linear relations(r>0.999).The standard recovery rate was between 97.45% and 103.4%.The differences of 4 constituents in Maidong from different producing areas were extremely significant(P<0.01).There was no significant difference of 4 constituents among different samples from the same area.Conclusion: The new established method for determination of representative components in Maidong can be applied to quality evaluation and geoherbalism analysis for Maidong.
