基于药材性状特征和含量测定分析干燥温度对麦冬质量的影响

目的:考察干燥温度与麦冬质量的相关性,为制定确定麦冬干燥工艺最佳参数提供理论支持。方法:单因素考察不同干燥温度条件下麦冬药材质量的变化特征,对麦冬性状进行比较分析;采用中国药典方法对浸出物含量进行测定,采用紫外分光光度法对多糖含量进行测定;采用HPLC-ELSD对麦冬皂苷D进行测定,以乙腈-水(48:52)为流动相;采用HPLC对麦冬甲基黄烷酮A、麦冬甲基黄烷酮B含量进行测定,以乙腈-水(58:42)为流动相,检测波长296nm;以乙腈-水为流动相梯度洗脱,在检测波长296nm条件下,建立HPLC指纹图谱。结果:麦冬药材质量随干燥温度升高而降低,干燥温度超过65℃时会导致麦冬药材颜色加深,有效成分含量显著下降。干燥从55℃上升到95℃浸出物含量下降20.5%,多糖含量下降17.0%,麦冬皂苷D含量降低41.8%,麦冬甲基黄烷酮A降低19.1%,麦冬甲基黄烷酮B降低13.5%。结论:干燥温度和麦冬药材质量呈负相关,综合考虑人工、时间和能耗等因素,设定60℃为麦冬最佳干燥温度参数。

Objective:To study the relationship of drying temperature and quality of Ophiopogonis Radix,so as to provide a basic theory to confirm the best drying technology parameters of Ophiopogonis Radix.Methods:The feature of quality change of Ophiopogonis Radix under the condition of different drying temperature was inspected by single factor test.The appearance character characteristics of Ophiopogonis Radix dried through different temperature were compared.The extracted content was measured by the method from ChP.Polysaccharide.Polysaccharide content was analyzed by ultraviolet spectrophotometry.Content of Ophiopogonin D was determined by HPLCELSD with acetonitrile-water(48:52)as mobile phase.Methylophiopogonanone A and Methylophiopogonanone B were evaluated by HPLC with acetonitrile-water(52:48)as mobile phase under the detection wavelength of 296 nm.The HPLC chromatogram was established using acetonitrile-wateras gradientelution mobile phase under the detection wavelength of 296 nm.Results:The quality of Ophiopogonis Radix was decreased with the increase of drying temperature.The color of the Ophiopogonis Radix would be darken and the content of effective constituents would be decreased when the drying temperature is above 65 ℃。The contents of extract,polysaccharide, Ophiopogonin D,Methylophiopogonanone A,Methylophiopogonanone B were respectively decreased by 20.5%, 17.0%,41.8%,19.1%,13.5% when the temperature increased from 55 ℃ to 95 ℃。Conclusion:Our results indicated a negative correlation between drying temperature and quality of Ophiopogonis Radix.Considering of labor cost, drying elapsed time and energy consumption,it is best to set the drying temperature parameters at 60 ℃.
