目的:运用近红外光谱技术(NIRS)测定不同产地野菊花总黄酮含量。方法:分别采集6个不同产地的野菊花样品,采用近红外漫反射光谱法采集不同产地野菊花近红外图谱;以芦丁为对照品,利用传统的分析方法紫外-可见分光光度法在502 nm处测定不同产地野菊花药材中总黄酮含量作为参考值,将近红外图谱与野菊花中总黄酮含量值进行关联,采用偏最小二乘法(PLS)建立定量分析模型,并对模型进行验证。结果:所建总黄酮定量分析模型的相关系数(R2)、校正均方差(RMSEC)、预测均方差(RMSEP)和交叉验证均方差(RMSECV)分别为0.98927、0.131、0.163和0.292;验证集样品的总黄酮含量近红外预测值的平均相对偏差为2.08%。结论:近红外光谱技术测定快速,操作简单,预测结果准确度高,可用于不同产地野菊花药材中总黄酮含量的快速测定。

Objective:To study the application of near infrared spectroscopy(NIRS) in the determination of the content of total flavonoids in Chrysanthemum of different habitats.Methods:Chrysanthemum of 6 different habitats were collected separately and the near infrared spectra of Chrysanthemum were obtained by Nicolet 6700 NIRS spectrometry.Using Rutin as the reference compound, the contents of total flavonoids in Chrysanthemum of different habitats were determined at the wavelength of 502 nm by the traditional analysis method of UV-Vis spectrophotometric method and used as reference values.The near infrared spectra and the contents of total flavonoids in Chrysanthemum of different habitats were correlated, and a quantitative analysis model was built and validated with partial least square(PLS) regression.Results:The correlation coefficient(R2), the root-mean-square error of calibration(RMSEC), the root-mean-square error of prediction(RMSEP) and the root-mean-square error of cross-validation(RMSECV) were 0.98927, 0.131, 0.163 and 0.292, respectively.The average relative deviation of the content of total flavonoids in validation set of samples predicted by NIRS was 2.08%.Conclusion:The method of NIR had the advantages of rapid determination, simple operation and high accuracy of prediction result, and can be used for rapid determination of total flavonoid content in Chrysanthemum of different habitats.
