Objective: To establish a RP-HPLC method for the simultaneous determination of eight hydrophilic and four lipophilic compounds including sodium danshensu, caffeic acid, isoferulic acid, salvianolic acid D, rosmarinic acid, lithospermic acid, salvianolic acid B, salvianolic acid A, 1, 2-dihydrotanshinquinone, cryptotanshinone, tanshinoneⅠ, tanshinoneⅡA in Salvia przewalskii Maxim.Methods: The HPLC separation was performed on a Shim-pack XR-ODSⅡ(75 mm×2.0 mm, 2.2μm) column at 30℃.The mobile phases consisted of acetonitrile-0.12% formic acid solution with gradient elution at a flow rate of 0.2 mL·min-1.The UV detection wavelength was 270 nm, the injection volume was 4μL.Results: The complete separation was obtained within 50 minutes for the 12 compounds.The linear ranges of sodium danshensu, caffeic acid, isoferulic acid, salvianolic acid D, rosmarinic acid, lithospermic acid, salvianolic acid B, salvianolic acid A, 1, 2-dihydrotanshinquinone, cryptotanshinone, tanshinoneⅠ, tanshinoneⅡA were 0.22-22.3μg·mL-1(r=0.9998, n=6), 0.05-4.91μg·mL-1(r=0.9998, n=6), 0.18-18.25μg·mL-1(r=1.000, n=6), 0.28-28.40μg·mL-1(r=0.9999, n=6), 13.50-1349.89μg·mL-1(r=0.9999, n=6), 0.23-23.22μg·mL-1(r=0.9997, n=6), 9.93-993.16μg·mL-1(r=0.9998, n=6), 0.04-3.87μg·mL-1(r=1.000, n=6), 0.81-81.33μg·mL-1(r=0.9998, n=6), 0.87-86.85μg·mL-1(r=0.9999, n=6), 0.67-67.47μg·mL-1(r=0.9994, n=6) and 1.01-101.12μg·mL-1(r=0.9998, n=6), respectively.The average recoveries(n=6) of sodium danshensu, caffeic acid, isoferulic acid, salvianolic acid D, rosmarinic acid, lithospermic acid, salvianolic acid B, salvianolic acid A, 1, 2-dihydrotanshinquinone, cryptotanshinone, tanshinoneⅠ, tanshinoneⅡA were 99.2%(RSD=1.7%), 99.7%(RSD=1.4%), 101.0%(RSD=2.0%), 99.7%(RSD=2.1%), 99.5%(RSD=1.9%), 101.2%(RSD=1.9%), 100.8%(RSD=1.7%), 100.7%(RSD=1.8%), 101.6%(RSD=1.5%), 100.7%(RSD=1.6%), 100.5%(RSD=1.4%) and 100.0%(RSD=1.7%), respectively.Conclusion: The method was validated to be simple, accurate, selective, and sensitive, which can be used for simultaneous determination of eight hydrophilic and four lipophilic components in Salvia przewalskii Maxim.and the quality control of Salvia przewalskii Maxim.and as the reference of the quality control of Salvia przewalskii Maxim.preparations.
