目的:建立HPLC法同时测定黄花列当中3个苯乙醇苷类化合物(毛蕊花糖苷、crenatoside、2'-乙酰毛蕊花糖苷)的含量。方法:采用Inertsil C18(4.6 mm×250 mm,5.0 μm)色谱柱,乙腈-0.1%磷酸水溶液(22:78)为流动相,流速1.0 mL·min-1,柱温25℃,检测波长330 nm。结果:毛蕊花糖苷、crenatoside和2'-乙酰毛蕊花糖苷在30 min内得到良好的分离,且进样量分别在0.104~5.20 μg(r=0.9998)、0.106~2.65 μg(r=1.0000)、0.058~1.15 μg(r=1.0000)范围内呈现良好线性;平均回收率(n=3)在96.24%~102.7%之间。13批黄花列当样品中毛蕊花糖苷、crenatoside、2'-乙酰毛蕊花糖苷3种成分的含量范围分别为1.40%~11.53%、1.35%~3.22%和0.15%~1.63%,其中含量高的2个成分是毛蕊花糖苷和crenatoside;不同产地的药材成分含量差异较大。结论:本方法操作简便,准确、重现性良好,可用于黄花列当药材和饮片的质量检测。

Objective:To develop an HPLC method for simultaneous determination of three phenylethanoid glycosides(acteoside,crenatoside,2'-acetylacteoside)in Orobanche pycnostachya.Methods:HPLC was performed on an Inertsil C18 analytical column(4.6 mm×250 mm,5.0 μm)with acetonitrile -0.1% aqueous phosphoric acid(22:78)as the mobile phase at a flow rate of 1.0 mL·min-1.The column temperature was 25℃, and the detection wavelength was set at 330 nm.Results:There was good linearity in the range of 0.104-5.20 μg(r=0.9998)for acteoside,0.106-2.65 μg(r=1.0000)for crenatoside,0.058-1.15 μg(r=1.0000) for 2'-acetylacteoside,respectively.The average recoveries(n=3)of acteoside,crenatoside and 2'-acetylacteoside were between 96.24%-102.7%.There were significant differences of the contents of three constituents in Orobanche pycnostachya from different sources.The content of acteoside is the highest,followed by crenatoside. The contents of acteoside,crenatoside and 2'-acetylacteoside in 13 batches of samples were1.40%-11.53%、1.35%-3.22% and 0.15%-1.63%,respectively.Conclusion:This method is proved to be simple,accurate and reliable,which can be used for quality inspection of Orobanche pycnostachya herbs and its decoction pieces.
