中药菊花基原特征成分的分析

目的:建立菊花药材中6种化合物(绿原酸、木犀草苷、3, 5-O-二咖啡酰基奎宁酸、木犀草素、山柰酚以及金合欢素)的HPLC含量测定方法及其指纹图谱检测方法,并对所收集到的市售5种药用菊花样品利用主成分分析获取能反映不同菊花样本间基原远近的关键成分。方法:含量测定部分采用YMCPark ODS-AQ C18(250 mm×4.6 mm, 5μm)色谱柱,以乙腈-0.01%磷酸为流动相梯度洗脱,流速0.8 mL·min-1,检测波长330 nm,柱温25℃;结合SPSS 16数据分析软件及SIMCA-P主成分分析软件对含量测定的数据进行分析,筛选出对菊花基原判定影响最大的特征成分组。结果:含量测定方法学验证结果良好,并且通过含量测定结果的数据分析,确立了与菊花基原判别最相关的特征成分组为木犀草素及木犀草苷。结论:本研究所建立的含量测定方法可以用于菊花药材中6种成分的定量分析,主成分分析所筛选出的基原远近的决定性成分因子,能大体上将不同来源的菊花样品按基原归类,为市售菊花基原鉴定提供参考依据。

Objective:To establish an HPLC method for determination of 6 kinds of compounds including chlorogenic acid,cynaroside,3,5-O-dicaffeoyl quinic acid,luteolin,kaempferol and acacetin in Chrysanthemum and evaluate the detection method of its fingerprint,so as to obtain the key components that can reflect the original distance of the collected 5 species of medicinal Chrysanthemum samples. Methods:Separation was performed on an YMC-Park ODS-AQ C18 chromatographic column(250 mm×4.6 mm, 5μm). A gradient elution was performed at the flow rate of 0.8 mL·min-1 with detection wavelength of 330 nm and column temperature at 25℃.SPSS 16 software and the SIMCA-P analysis procedure were used for data analysis to screen out the key components which affected the judgement of the original about Chrysanthemum. Results:Methodology validation of this content determination method was good.Through the analysis of the data on content determination results of Chrysanthemum, the most relevant component group for judging the origin of Chrysanthemum was identified as cynaroside and luteolin. Conclusion:This study had established a determination method for detecting the 6 kinds of ingredients whose quantities were obvious different due to the diversity of the types in Chrysanthemum. The decisive factor selected by principal component analysis can generally classify different sources of chrysanthemums.This identification method can provide reference for the original identification of commercially available Chrysanthemum.
