目的: 建立高效液相色谱法同时测定白背叶药材中葫芦巴苷Ⅱ、异夏佛托苷、夏佛托苷3个黄酮碳苷类成分含量. 方法 :采用Agilent EclipseXDB-C18色谱柱(4.6 mm×250 mm,5 μm),柱温25 ℃,流动相为甲醇-0.1%乙酸水溶液,梯度洗脱,流速1.0 mL · min-1,检测波长336 nm. 结果 :葫芦巴苷Ⅱ、异夏佛托苷、夏佛托苷3个成分的色谱峰均达到基线分离,其进样量分别在0.026 6~5.32、0.024 4~4.88、0.024 2~4.84 μg范围内线性关系良好(r=0.999 0),平均加样回收率(n=6)分别为97.3%、100.1%、103.0%;不同批号的白背叶药材葫芦巴苷Ⅱ、异夏佛托苷、夏佛托的含量分别为0.73~7.48、1.25~7.58、1.03~5.89 mg · g-1. 结论: 本文所建立的含量测定方法经方法验证适于白背叶药材的质量检测.

Objective: To establish an HPLC method for simultaneous determination of three flavone glycosides (vicenin Ⅱ,isoschaftoside and schaftoside) in Mallotus apelta. Methods : HPLC was performed on an Agilent Eclipse XDB-C18 analy tical column (4.6 mm×250 mm,5 μm) at 25 ℃ with methanol and 0.1% acetic acid as the mobile phase by gradient elution,the flow rate was 1.0 mL · min-1 and the detection wavelength was 336 nm. Results :The baseline se paration of these three components was achieved; the linear ranges of vicenin Ⅱ,isoschaftoside and schaftoside were 0.026 6-5.32,0.024 4-4.88,and 0.024 2-4.84 μg,respectively (r=0.999 0); the average recoveries(n=6) were 97.26%,100.1,and 103.0%,respectively.The content ranges of vicenin Ⅱ,isoschaftoside and schaftoside were 0.73-7.48,1.25-7.58 and 1.03-5.89 mg · g-1 for Mallotus apelta from different batch number. Conclusion: The established method of determination can be used for quality control of Mallotus apelta.
