光谱成像指纹图谱在药用珍珠鉴定中的应用研究

目的: 建立珍珠粉的光谱成像指纹图谱,为其质量控制提供新的方法。方法: 应用电可控液晶滤光光谱成像装置,测定8种不同市售来源的药用珍珠粉和3种伪品样本,从成像光谱立方体中提取特征光谱,构建其指纹图谱和质量等级分类。基于欧氏距离计算,根据聚类法解析其指纹图谱。结果: 样品1#~5#的特征光谱曲线与6#~11#显著不同,聚类分析提示样品6#~11#为同一类群、1#~5#为其他类群。质量等级分类示样品8#、11#为A级,6#、7#、9#、10#为B级,4#、5#为C级。光谱成像指纹图谱的鉴定结果与对照试验中性状、理化及紫外的鉴定结果相符合,即6#~11#为珍珠来源、样品1#~5#为非珍珠来源。结论: 光谱成像分析技术可用于珍珠粉指纹图谱的构建,能快速准确地鉴别不同品系珍珠类产品和辨别真伪,在品质评价和质量鉴定上较传统法更为简单方便,有效稳定,其快速鉴别特点为今后应用于中药材质量控制提供了借鉴。

Objective: To establish spectral imaging fingerprints of pearl powder,and provide a new approach for quality control. Methods: Eight kinds of commercial Chinese herbal pearl powder of different sources and three fake and shoddy samples were measured by electric controllable liquid crystal filtration spectral imaging device. Characteristic spectra were extracted from the imaging spectral cube,the fingerprints were constructed to make preliminary discrimination and quality grading. Based on calculation of euclidean distance,the fingerprints were interpreted on the basis of clustering analysis. Results: The character fluorescence spectral curves of samples 1#-5# were obviously different from samples 6#-11#. The cluster analysis distributed samples 6#-11# to the same class group and samples 1#-5# to the other class group. Quality grade classification showed that samples 8#,11# were in grade A,samples 6#,7#,9#,10# were in grade B,and samples 4#,5# were in grade C. The identification results of the spectral imaging fingerprint were consistent with the results of morphological,physico-chemical and ultraviolet identification,demonstrating that samples 6#-11# were pearl origin while samples 1#-5# were not. Conclusion: Spectral imaging analytical technique is available for imaging processing of pearl powder fingerprint,and the method can rapidly and accurately identify and differentiate pearl powder collected from different sources. It proves to be more simple,convenient,robust and effective than traditional methods in quality evaluation and identification. Its character of rapid identification provides reference for further application of spectral imaging techniques in quality control of Chinese crude drugs.
