藏雪莲多糖对UVB辐射HaCaT细胞氧化损伤的保护作用

目的: 探讨藏雪莲(Saussurea tridactyla Sch.-Bip.)多糖能否减轻中波紫外线(ultraviolet B,UVB)辐射后人角质形成细胞(HaCaT cell)的氧化损伤。方法: 体外培养HaCaT细胞,分为低辐射组(30 mJ·cm-2,辐射1 h,L-UVB)、高辐射组(60 mJ ·cm-2,辐射1 h,H-UVB),且L-UVB组、H-UVB组又分别分为对照组、辐射组(UVB组)、低剂量(10 mg·mL-1)藏雪莲多糖组(低剂量组)、高剂量藏雪莲多糖(40 mg·mL-1)组(高剂量组)。藏雪莲多糖于UVB辐射前1 h加入细胞培养基中,照射后18 h,收集细胞。采用台盼蓝染色观察细胞形态及死亡情况,(MTS)法检测细胞增殖,酶标法检测超氧化物歧化酶(SOD)活力,谷胱甘肽(GSH)含量,细胞丙二醛(MDA)含量,过氧化氢酶(CAT)活力。结果: L-UVB组与H-UVB组中低、高剂量藏雪莲多糖组与UVB组比较,台盼蓝染色后,形态明显恢复,细胞蓝染数量明显下降,细胞死亡碎片及脱片现象明显改善,MTS法检测细胞增殖增高,酶标法检测SOD活力,GSH含量,CAT活力增高,MDA含量下降,且与辐射组比较差异均具有统计学差异(P<0.05)。结论: 藏雪莲多糖对UVB辐射HaCaT细胞的氧化损伤有保护作用。

Objective: To study whether Saussurea tridactyla Sch.-Bip.polysaccharide could reduce oxidative damage caused by ultraviolet B(UVB)on HaCaT cells. Methods: HaCaT cells were cultured in vitro and divided into low radiation group(30 mJ·cm-2,radiation 1 h,L-UVB)and high radiation group(60 mJ·cm-2,radiation 1 h,H-UVB).L-UVB group and H-UVB group were respectively divided into control group,UVB radiation group(UVB group),Saussurea tridactyla Sch.-Bip.polysaccharide low dose group(10 mg·mL-1,low dose group),and Saussurea tridactyla Sch.-Bip.polysaccharide high dose group(40 mg·mL-1,high dose group).Before 1 h of UVB irradiation,Saussurea tridactyla Sch.-Bip.polysaccharide was added into the culture medium.After 18 h,cells were collected.The morph and death of cells were observed by trypan blue staining.Cell proliferation was detected by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium,inner salt(MTS).Superoxide dismutase(SOD)activity test kit,glutathione(GSH)content test kit,malondialdehyde(MDA)content test kit and catalase(CAT)activity test kit were used to detect SOD activity,GSH content,MDA content,and CAT activity. Results: In L-UVB group and H-UVB group,the morphology of cells was restored;the number of dead cells was reduced;fragments of dead cells and chip phenomenon were obviously improved in low and high dose of Saussurea tridactyla Sch.-Bip.polysaccharide groups compared with UVB group by trypan blue staining.The cell proliferation was increased by detected by MTS.SOD activity,GSH content,and CAT activity were increased,MDA content was decreased,and had statistically difference compared with UVB group(P<0.05). Conclusion: Saussurea tridactyla Sch.-Bip.polysaccharide had the protective effect against oxidative damage by UVB irradiation on HaCaT cells.
