微波消解-ICP-MS测定人血白蛋白中痕量元素

目的: 建立微波消解电感耦合等离子体质谱测定人血白蛋白中痕量元素的方法. 方法: 人血白蛋白经过微波消解,并对电感耦合等离子体质谱仪器工作参数进行优化,采用标准模式和动能歧视模式同时对Cr、Mn、Fe、Co、Ni、Cu、Zn、Ag、Cd、Sb、Cs、Ba、Tl、Pb 14种元素进行检测,RF功率1.4 kW,载气流速1.07 mL·min-1,辅助气流速1.20 mL·min-1,同心雾化器,蠕动泵转速40 r·min-1.采用混合内标(74Ge、103Rh、115In、175Lu、209Bi)进行校正. 结果: 痕量元素的检测限为0.01~0.21 μg·L-1,方法精密度为0.1%~5.5%,准确度为81.9%~116.8%,回收率为96.0%~113.0%.用该方法对国家标准物质头发成分GBW07601(GSH-1)进行分析测定,各微量元素测定值在标准值的范围内. 结论: 本方法适用于人血白蛋白中痕量元素的测定.

Objective: To develop a method for determining trace elements in human albumin by using microwave digestion-ICP-MS. Methods: Under optimized conditions of ICP-MS the samples(Cr,Mn,Fe,Co,Ni,Cu,Zn,Ag,Cd,Sb,Cs,Ba,Tl,Pb)were analyzed by standard(STD)mode and the kinetic energy discrimination(KED)mode after microwave digestion.The RF power was 1.4 kW,the flow rate of carrier gas was 1.07 mL·min-1 and the flow rate of auxiliary gas was 1.20 mL·min-1.The concentric nebulizer was used,and the rotation rate of peristaltic pump was 40 r·min-1.The mixed internal standards(74Ge,103Rh,115In,175Lu,209Bi)were used for calibration in trace element analysis. Results: The analytical results showed that the determination limits of the trace elements were in the range of 0.01-0.21 μg·L-1.The precision of the method ranged from 0.1% to 5.5%.The accuracy was in the range of 81.9%-116.8%.The recovery rates of the studied elements were from 96.0% to 113.0%.The national standard reference material GBW07601(GSH-1)was analyzed by the described method and the measured element values accorded with the standard values. Conclusion: The method could be used for determining trace elements in human albumin.
