HPLC测定亚贡叶中亚贡二萜酸A的含量

目的: 建立高效液相色谱法亚贡叶中亚贡二萜酸A的含量. 方法: 采用Agilent ZORBAX SB-C18(4.6 mm×150 mm,5 μm)色谱柱,以乙腈-0.5%磷酸水溶液(35:65)为流动相,流速1.0 mL·min-1,检测波长203 nm,柱温30℃. 结果: 亚贡二萜酸A的线性范围为0.064~1.028 μg,r=0.999 9;加样回收率为99.6%,RSD为1.5%;批号为20101001、20101015、20101020的3批样品中亚贡二萜酸A含量分别为0.531、0.654、0.294 mg·g-1. 结论: 不同来源的亚贡叶的质量差异较大;方法学验证结果表明本法适用于亚贡叶的质量控制.

Objective: To establish an HPLC method for determination of smallanthaditerpenic acid A in leaves of Smallanthus sonchifolius. Methods: The determination was performed by HPLC with Agilent ZORBAX SB-C18(4.6 mm×150 mm,5 μm) column using acetonitrile-0.5% phosphate solution(35: 65) as the mobile phase,the flow rate was 1.0 mL·min-1,the detection wavelength was 203 nm and the column temperature was 30℃. Results: There was a good linear relationship with a range of 0.064-1.028 μg for smallanthaditerpenic acid A,r=0.999 9;the average recovery rate of smallanthaditerpenic acid A was 99.6%,RSD was 1.5%. The contents of Smallanthus sonchifolius samples were 0.531 mg·g-1 for lot No.20101001,0.654 mg·g-1 for lot No.20101015,and 0.294 mg·g-1 for lot No.20101020. Conclusion: The quality differences of Smallanthus sonchifoliu leaves from different sources are great;the results of method validation show that the method can be used to control quality of Smallanthus sonchifolius.
