迷迭香药材HPLC特征图谱分析研究

目的: 建立迷迭香的指纹图谱分析方法,并比较迷迭香不同部位(茎、叶)的指纹图谱,为评价迷迭香药材质量提供参考.方法: 采用HPLC法测定,色谱柱为Alltima C18柱(250 mm×4.6 mm,5 μm);流动相为乙腈-0.1%甲酸(梯度洗脱),流速1 mL·min-1,检测波长280 nm,柱温为25 ℃.采用国家药典委员会中药色谱指纹图谱相似度评价系统软件(2004 A版),分析迷迭香药材指纹图谱的共有模式及样品的相似度.结果: 建立了迷迭香药材HPLC指纹图谱共有模式,标定了9个共有峰.9批6个不同产地迷迭香的HPLC特征谱图相似度均在0.98以上.在不同时间对样品进行测定,各特征峰的保留时间的RSD小于0.02%,相对峰面积的RSD小于0.2%,结果表明供试品溶液在24 h内稳定.对3批迷迭香的茎叶进行特征图谱分析,均以叶的相似度为高,其相似度在0.98以上.结论: 本研究发现,迷迭香茎叶所含成分基本相同,但叶中成分的量基本高于茎.所建立的方法经方法学验证,可为迷迭香药材的质量控制提供依据.

Objective: To establish a method of fingerprint analysis on Rosmarinus officinalis by HPLC, and compare the variability of the different parts, including stems and leaves, so as to provide reference for quality evaluation of Rosmarinus officinalis.Methods: The Alltima C18 column(250 mm×4.6 mm, 5 μm)was used with acetonitrile-0.1% formic acid solution(gradient elution)as the mobile phase at the flow rate of 1 mL·min-1 and the detection wavelength of 280 nm, and the column temperature was 25 ℃.Results: HPLC-fingerprint of Rosmarinus officinalis from different areas was established, which contained 9 common peaks.The similarity of the samples of 9 batches of Rosmarinus officinalis from 6 producing areas was higher than 0.98.Relative standard deviation(RSD)for relative retention times of various specific peaks obtained at different time points of sample introduction was less than 0.02%, and RSD for the relative peak areas was less than 0.2%.The results obtained proved that the sample was stable within 24 h.The fingerprints of the stem and leaves of Rosmarinus officinalis from 3 batches were analyzed and showed that the similarity in leaves was higher which was more than 0.98.Conclusion: The study results showed that the components in stems and leaves of Rosmarinus officinalis were the same, but the content in leaves was higher than that in stems.The method by the methodological validation can be a reference for the quality control of Rosmarinus officinalis.
