溴化十六烷基三甲铵琼脂培养基促生长能力的评价分析

目的: 通过对铜绿假单胞菌在溴化十六烷基三甲铵琼脂培养基中生长性能差异的分析,建立培养基的评价分析方法. 方法: 利用96孔板设置不同培养基处方(3种胨源×2种培养基配方)、溴化十六烷基三甲铵浓度(6水平)和铜绿假单胞菌浓度(4水平)的组合,分析在各种不同的组合条件下铜绿假单胞菌的生长情况. 结果: 不同溴化十六烷基三甲铵琼脂促生长能力的差异主要由胨源和配方所决定;USP/EP收载的溴化十六烷基三甲铵琼脂配方优于目前中国药典收载的配方. 结论: 多因素多水平的微量分析方法可较好地揭示影响培养基促生长能力的诸因素,为培养基配方的筛选开拓新思路.

Objective: To develop a novel method for evaluation of the medium by analyzing differences in the growth property of Pseudomonas aeruginosa on cetrimide agar. Methods: Combinations of different media(3 peptones×2 medium formulas)with cetrimide concentrations(6 levels)and Pseudomonas aeruginosa concentrations(4 levels)were set up on the 96-well plate.And the growth promoting property of Pseudomonas aeruginosa under different combination conditions was analyzed. Results: Peptone source and medium formula were the main determinants for the growth property differences of Pseudomonas aeruginosa on cetrimide agar.The cetrimide agar formula in USP/EP was better than that in ChP 2010. Conclusion: Trace analysis with multiple factors and multiple levels could well exhibit all the factors referring to the growth promotion property of medium and provide new thoughts for the selection of medium formula.
