HPLC法同时测定暴马子皮中紫丁香苷、橄榄苦苷的含量

目的:建立测定暴马子皮药材中2个成分(紫丁香苷、橄榄苦苷)的高效液相色谱分析方法。方法:采用Shiseido CAPCELL PAK-C18(250 mm×4.6 mm,5 μm)色谱柱,流动相为0.1%甲酸水溶液(A)-乙腈(B),梯度洗脱(0~45 min,5%B→40%B),流速1.0 mL·min-1,DAD检测器,检测波长280 nm,柱温25 ℃。结果:紫丁香苷、橄榄苦苷线性范围分别为0.25~0.74 μg(r=0.9987)和0.9~13.1 μg(r=1.0000),平均加样回收率(n=6)分别为97.5%(RSD=1.3%)和98.2%(RSD=1.1%)。结论:不同来源的暴马子皮药材中紫丁香苷、橄榄苦苷含量均存在显著性差异。本方法操作简单快速,重复性好,为暴马子皮药材的质量控制研究奠定了良好的基础。

Objective: To develop an HPLC method for the determination of syringin and oleuropein in Syringae Cortex. Methods: Determination was carried out on a Shiseido CAPCELL PAK-C18 (250 mm×4.6 mm,5 μm)column.The mobile phase was composed of water solvent containing 0.1% formic acid(A)and acetonitrile(B)with gradient elution(0-45 min,5%B→40%B)at the flow rate of 1.0 mL·min-1,the detection wavelength was 280 nm for the DAD detector and the column temperature was set at 25 ℃. Results: The calibration curves were linear in the ranges of 0.25-0.74 μg for syringin(r=0.9987)and 0.9-13.1 μg for oleuropein(r=1.0000).The average recoveries(n=6)of syringin and oleuropein were 97.5%(RSD=1.3%)and 98.2%(RSD=1.1%)respectively. Conclusion: The yields of the two components in different origins exhibit significant differences.This method is simple,quick and repeatable,which lays a good foundation for the quality control of Syringae Cortex.
