线性回归色谱峰定位法在射干药材多组分同时测定中的应用

目的:将线性回归色谱峰定位法应用于射干药材多种药效组分的一测多评研究中,提高一测多评方法的准确度及可行性。 方法:以射干药材中6个药效组分(射干苷、野鸢尾苷、鸢尾黄素、野鸢尾黄素、次野鸢尾黄素、白射干素)为研究对象,以射干苷为对照品计算其他各组分的相对校正因子;以射干苷及次野鸢尾黄素对照品建立线性方程,进行待测成分色谱峰定位;考察线性回归法在6根不同色谱柱上对待测组分色谱峰定位的结果,以考察方法的适用性及耐用性;与相对保留时间及保留时间差定位法进行比较,以考察线性回归色谱峰定位法的优越性;将用校正因子计算得到的结果与外标法测定结果进行比较,以考察方法的准确性。 结果:采用线性回归法进行色谱峰定位时,保留时间理论值与测定值绝对误差很小,优于相对保留时间定位法;提高了色谱峰定位的准确性;实验中建立的校正因子重现性良好,4批药材中6个组分含量的计算值与外标法实测值之间通过夹角余弦法检验无显著性差异。 结论:线性回归色谱峰定位法的应用可使多组分同时测定方法更具合理性和可行性。

Objective: To apply a new method of linear regression chromatographic peak positioning in the quantitative analysis of multicomponents by single marker (QAMS),accomplishing a simultaneous assay of Rhizoma Belamcandae.The new method can improve the accuracy and the feasibility of the assay. Methods: Six effective components (tectoridin,iridin,tectorigenin,irigenin,irisflorentin,dichotomitin) were used as the research object,and tectoridin was selected as the reference substance to validate other components;tectoridin and irisflorentin were used to establish the linear equations for positioning of the chromatographic peaks;the feasibility and applicability of this method were checked on six different chromatographic columns;the superiority of the linear regression chromatographic peak positioning was investigated by comparing the localization method of the relative retention time and the retention time difference;the accuracy of the method was evaluated by comparison of the quantitative results between external standard method and QAMS. Results: The absolution error between the theoretical value and the measured value of the retention time was very small when using the linear regression method to position the chromatographic peaks,which was more accurate than the relative retention time positioning method;the accuracy of chromatographic peak positioning was improved;the correction factor established in the experiment had good reproducibility;there was no significant difference between the calculated values of 6 components in 4 batches and the measured values in the external standard method in angular cosine test. Conclusion: The new method of linear regression chromatographic peak positioning increased the reasonability and feasibility of simultaneous determination of multicomponents.
