桃儿七根茎HPLC指纹图谱研究

目的:采用HPLC法建立桃儿七根茎药材的指纹图谱,并测定不同产地11批药材中鬼臼毒素的含量。方法:采用Wondasil C18(4.6 mm×150 mm,5 μm),以0.1%甲酸水溶液(pH 3.2)为流动相A,甲醇为流动相B,梯度洗脱(0.01 min,35%B;25 min,47%B;85 min,55%B;95 min,60%B;97 min,35%B;100 min,35%B;100.01 min,STOP),流速0.8 mL·min-1,检测波长290 nm,柱温25 ℃,进样量10 μL。结果:以鬼臼毒素为参照峰确立了36个共有峰,建立了桃儿七HPLC指纹图谱,共有峰面积达到总峰面积90%以上,11批次药材相似度均在0.9以上;各批次药材中鬼臼毒素含量在13.5~36.9 mg·g-1间。结论:建立的HPLC指纹图谱方法可同时用于含量测定,该方法操作简单,重复性及稳定性良好,可反映桃儿七药材质量。

Objective: To establish the chromatographic fingerprint of the rhizome of Sinopodophyllum hexandrum (Royle) Ying by HPLC method,and perform the quantitative analysis of podophyllotoxin. Methods: The 11 batches of samples from different producing areas were analyzed by HPLC with a Wondasil C18 column (4.6 mm×150 mm,5 μm),gradiently eluted(0.01 min,35%B;25 min,47%B;85 min,55%B;95 min,60%B;97 min,35%B;100 min,35%B;100.01 min,STOP)with mobile phase A (water containing 0.1% methanoic acid pH=3.2)-mobile phase B (methanol) at 25 ℃,and monitored with UV detector at 290 nm.The flow rate was 0.8 mL·min-1,and the sample volume injected was 10 μL. Results: Thirty-six mutual peaks were selected as the fingerprint peaks with podophyllotoxin as the reference compound.The rate of common peak area to the total peak area was higher than 90%,and the similarities of the correlation coefficients were higher than 0.9.The content of podophyllotoxin in different batches ranged from 13.5 to 36.9 mg·g-1. Conclusion: The method is simple,reproducible,and stable,which can serve as a scientific and effective way for content determination and quality control of S.hexandrum.
