大菟丝子生品和盐炙品HPLC-UV特征图谱及7个有机酸含量变化研究

目的:建立大菟丝子生品和盐炙品的特征图谱,探讨大菟丝子炮制前后化学成分的变化规律,对其中的新绿原酸、绿原酸、隐绿原酸、咖啡酸、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸7个有机酸类成分进行定量分析比较。方法:采用HPLC-UV法,选用Odyssil C18 色谱柱(4.6 mm×250 mm,5 μm),流动相为乙腈(A)-0.05%磷酸溶液(B),梯度洗脱(0~5 min,5%A→13%A;5~20 min,13%A→13%A;20~25 min,13%A→20%A;25~45 min,20%A→40%A),流速1 mL·min-1,检测波长325 nm,柱温35 ℃;用国家药典委员会“中药色谱指纹图谱相似度评价系统2004A”处理分析。结果:指标性成分在测定范围内与峰面积线性关系良好(r≥0.9970);不同来源的大菟丝子生品或盐炙品间的特征图谱相似度较高,但炮制前后指标性成分含量差异显著,呈现炮制后新绿原酸、隐绿原酸、3,4-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸呈倍数增长,而绿原酸、咖啡酸、3,5-二咖啡酰奎宁酸呈倍数下 降的趋势。指纹图谱评价生品(S1~S8)、盐炙品(S9~S16)、药房样品(S17~S25)特征图谱相似度发现生品与盐炙品间差异相对较大,而盐炙品与药房样品的相似度最小。结论:采用特征图谱结合7个有机酸指标性成分含量测定的方法可以较好地控制大菟丝子生品、盐炙品的质量,并可用于生、盐炙品的区分。

Objective: To develop the HPLC-UV specific chromatograms of Cuscutae Japonicae Semen before and after salt processing,to explore the regularity of component changes in processing of Cuscutae Japonicae Semen,and conduct quantitative analysis and compare neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,coffeic acid,3,4-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid,4,5-dicaffeoylquinic acid. Methods: HPLC-UV was used,and the separation was performed on an Odyssil C18 column (4.6 mm×250 mm,5 μm),the mobile phase was acetonitrile (A)-0.05% phosphoric acid (B) in gradient elution (0-5 min,5%A→13%A;5-20 min,13%A→13%A;20-25 min,13%A→20%A;25-45 min,20%A→40%A),at the flow rate of 1.0 mL·min-1,the column temperature was set at 35 ℃ and the detection wavelength was at 325 nm.The data were analyzed with similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine (2004A). Results: The linear correlation of major components was good (r≥0.9970).HPLC-UV specific chromatograms of Cuscuta Japonica Semen from different areas were highly similar,while the differences were obvious in contents of major components,and the contents of neochlorogenic acid,cryptochlorogenic acid,3,4-dicaffeoylquinic acid,4,5-dicaffeoylquinic acid increased exponentially after processing;on the contrary,the contents of chlorogenic acid,coffeic acid,and 3,5-dicaffeoylquinic acid decreased exponentially.The similarity test by fingerprint found the difference between raw products and salt products was relatively greater,while the difference between raw products and pharmacy products was the minimal. Conclusion: The HPLC-UV specific chromatogram and content determination of seven organic acids can be used for the quality control and distinction of Cuscutae Japonicae Semen before and after salt processing.
