薄层色谱和高效液相色谱法鉴别真伪蜂蜜的方法研究

目的: 建立检查药用蜂蜜真伪的色谱检测方法,为中国药典蜂蜜标准的修订提供依据。 方法: 试样经活性炭-硅藻土柱处理,采用薄层色谱法和高效液相色谱-蒸发光散射检测器检测法检测,色谱分离选用Prevail Carbohydrate ES 5u色谱柱(4.6 mm×250 mm,5 μm),流动相为乙腈-水,梯度洗脱(0~40 min,75%A→55%A,25%B→45%B),检测寡糖成分的检出。 结果: 以麦芽糖系列寡糖对照,检测结果显示,天然蜂蜜不含有五糖以上的寡糖;而果葡糖浆、麦芽糖浆及其蜂蜜的掺伪品均可检出大于麦芽五糖的寡糖。 结论: 可通过检测寡糖的存在与否鉴别蜂蜜的真伪。

Objective: To establish methods for the determination of the "authenticity" of honey,and provide basis for the standard revision of Chinese Pharmacopoeia. Methods: The sample was processed by a column packed with activated carbon and kieselguhr,separated by TLC and HPLC-ELSD methods.The analysis was performed on Prevail Carbohydrate ES 5u chromatographic column(4.6 mm×250 mm,5 μm)with a gradient elution program using mobile phase of acetonitrile and water(0~40 min,75%A→55%A,25%B→45%B).The presence of oligosaccharides,molecular weight larger than maltopentaose,was detected in the sample. Results: No oligosaccharide was detected in the batches of natural honey samples,which was found in high fructose corn syrup,maltose syrup,and the honey adulteration products. Conclusion: Oligosaccharide could be adopted as a marker for the authenticity of natural honey.
