UPLC-MS/MS法测定吸毒者头发中10种毒品代谢物的含量

目的:建立快速、准确的UPLC-MS/MS方法测定人头发中残留的海洛因、冰毒、摇头丸、苯环己哌啶、美沙酮、可尤因、氯胺酮等10种毒品。方法:头发样品经清洗、剪碎,甲醇-乙腈-2 mmol·L-1甲酸铵溶液(25:25:50),超声提取,5种同位素内标物,采用Zorbax ECLIPSE Plus C18(2.1 mm × 50 mm,1.8 μm)色谱柱,0.1%甲酸乙腈-0.1%甲酸水为流动相,流速 0.35 mL·min-1梯度洗脱,质谱采用ESI+离子源,MRM监测模式,检测6-乙酰基吗啡、吗啡、可待因、甲基安非他明、亚甲二氧基甲基苯丙胺、亚甲二氧基乙基苯丙胺、亚甲二氧基苯丙胺、苯环己哌啶、美沙酮、苯甲酰爱康宁、可卡因、去甲可卡因、古柯乙烯、去甲氯胺桐、氯胺酮、6-乙酰基吗啡,16种化合物的定量离子对和定性离子对。结果:16种化合物在2~1000 ng·mL-1浓度范围内线性良好,日间和日内精密度RSD均小于14.6%,回收率在83.3%~110%。结论:本方法可用于吸毒者头发中残留的10种毒品代谢物的含量测定。

Objective: To develop a rapid and accurate method for the determination of 10 drugs resided in human hair by UPLC-MS/MS,whith were heroine,ice,ecstasy,pcp,metadone,cocaine,ketamine,et al. Method: Hair samples were decontaminated and cut,followed by ultrasonic extraction with a solution consisted of methanol:acetonitrile:2 mmol·L-1 ammonium formate solution(25:25:50). Using 5 kinds of isotope standard as the internal standard,the samples were separated on a Zorbax ECLIPSE Plus C18 (2.1 mm×50 mm,1.8 μm)column,eluted at the flow rate of 0.35 mL ·min-1 by 0.1% formic acid-acetonitrile and 0.1% formic acid-water. MS instrument equipped with ESI+ ion source and MRM mode was used to detect the quantitative and qualitative ion pairs of 16 compounds,whith were 6-acetylmorphine,morphine,codeine,methamphetamine,amphetamine,MDMA,MDA,phencyclidine,methadone,benzoylecgonine,cocaine,norcocaine,cocaethylene,norkeamine,ketamine,6-acetylmorphine-D6. Results: The 16 compounds showed a good linearity in the concentration range of 2-1000 ng·mL-1,RSD of the intra-and inter-day precision was less than 14.6%,and the recovery was in the range of 83.3%-110%. Conclusion: This method is suitable for the quantitative determination of the metabolites of 10 drugs in the hair of drug abusers.
