目的:建立藕节药材中3-表白桦脂酸含量测定及HPLC指纹图谱分析的方法。方法:采用Waters Xbridge C18色谱柱(250 mm×4.6 mm,5 μm),以乙腈-0.05%磷酸水溶液为流动相,流速1.0 mL·min-1,检测波长203 nm,柱温35℃。结果:测定了不同产地藕节药材中3-表白桦脂酸的含量,并建立了藕节药材的HPLC指纹图谱,在指纹图谱中共有11个共有峰,并指认了3-表白桦脂酸,指纹图谱的相似度均在0.942~0.999之间,各色谱峰分离效果良好,符合指纹图谱检测要求。结论:该方法较好地反映了藕节药材中的化学成分信息,且操作简单,重复性和耐用性好,为进行藕节药材的质量评价提供了参考。

Objective: To establish a method for the determination of 3-epibetulinic acid and the analysis of the chemical fingerprint of Nodus Nelumbinis Rhizomatis by HPLC. Methods: The Waters Xbridge C18 column(250 mm×4.6 mm,5 μm) was used with acetonitrile-0.05% phosphoric acid aqueous at a flow rate of 1.0 mL·min-1,the monitoring wavelength was 203 nm and the column temperature was 35℃. Results: The contents of 3-epibetulinic acid in Nodus Nelumbinis Rhizomatis collected from different producing areas were determined,and the HPLC fingerprint of Nodus Nelumbinis Rhizomatis was established. Eleven common peaks were selected as characteristic peaks in the chromatogram of Nodus Nelumbinis Rhizomatis,and the fourth peak was identified as 3-epibetulinic acid. The similarities varied from 0.942 to 0.999. Each peak in the fingerprint was separated well with the above condition,and conformed to requirements of fingerprint detection. Conclusion: The method can display the chemical information of Nodus Nelumbinis Rhizomatis well.It is simple,reliable and durable,which can be used for the quality control of Nodus Nelumbinis Rhizomatis.
