RP-HPLC法同时测定芎菊上清丸中绿原酸、盐酸小檗碱和黄芩苷的含量

目的：建立RP-HPLC法同时测定芎菊上清丸中绿原酸、盐酸小檗碱和黄芩苷的含量。方法：采用DIKMA Platisil ODS色谱柱（250 mm×4.6 mm，5 μm），柱温30℃，以甲醇（A）-0.1%磷酸水溶液（B）为流动相，梯度洗脱（0～5 min，40%A;5～20 min，40%A→60%A；20～30 min，60%A），流速1 mL·min^-1，变换波长检测（327 nm，绿原酸;345 nm，小檗碱；278 nm，黄芩苷）。结果：绿原酸、盐酸小檗碱和黄芩苷的线性范围分别为0.11～2.2 μg（r=0.9997）、0.021～0.42 μg（r=0.9999）、0.03～0.6 μg（r=0.9998），平均回收率在98.45%和99.07%之间。结论：该法检测效率高，专属性强，重复性好，可用于芎菊上清丸中绿原酸、盐酸小檗碱和黄芩苷的含量测定及其质量控制。

Objective: To establish a RP-HPLC method for simultaneous determination of chlorogenic acid,berberine hydrochloride and baicalin in Xiongju Shangqing pills. Methods: The separation was performed on a DIKMA Platisil ODS column(250 mm ×4.6 mm,5 μm) with gradient elution(0-5 min,40%A;5-20 min,40%A→60%A;20-30 min,60%A) using methanol(A) and 0.1% phosphoric acid aqueous solution(B) as mobile phase at a flow rate of 1 mL·min^-1. The detection wavelengths were 327 nm for chlorogenic acid,345 nm for berberine and 278 nm for baicalin,respectively. Results: Linear ranges of chlorogenic acid,berberine hydrochloride and baicalin concentrations were 0.11-2.2 μg(r=0.9997),0.021-0.42 μg(r=0.9999),0.03-0.6 μg(r=0.9998),respectively. Their average recoveries fell between 98.45% and 99.07%. Conclusion: The RP-HPLC method established can be used to determine the contents of chlorogenic acid,berberine hydrochloride and baicalin in Xiongju Shangqing pills,which may also be applied as a standard for the quality control of Xiongju Shangqing pills.
