MEKC-DAD同时测定黄芩中6个黄酮类成分的含量

目的: 建立胶束电动毛细管色谱二极管阵列检测法(MEKC-DAD)同时测定不同批次黄芩及其炮制品中黄芩苷、黄芩素、汉黄芩苷、汉黄芩素、千层纸素A及芹菜素含量。 方法: 以40 mmol·L-1磷酸氢二钠-15 mmol·L-1 硼砂-40 mmol·L-1十二烷基硫酸钠-15%乙腈-7.5%丙醇(pH 8.4)为电泳介质,未涂渍标准熔融石英毛细管(75 μm×64.5 cm,有效长度56 cm)为分离通道,分离电压为20 kV,检测波长为280 nm,毛细管温度为25 ℃,压力进样为5 kPa×6 s。 结果: 6个黄酮的浓度与峰面积的线性关系良好(r>0.9990) ;加样回收率为96.20%~103.6%。用此法测定了黄芩中6个黄酮类成分的含量,得到满意结果。 结论: 该方法简单、准确,重复性较好,可用于黄芩药材内在质量的评价和控制。

Objective: To establish a method for simultaneous determination of baicalin, baicalein, wogonoside, wogonin, oroxylin A and apigenin in Scutellariae Radix of different batches and its processed products by micellar electrokinetic chromatography with diode array detection(MEKC-DAD). Methods: The buffer solution (pH 8.4) of 40 mmol·L-1 sodium hydrogen phosphate-15 mmol·L-1 sodium borate-40 mmol·L-1 SDS-15% acetonitrile-7.5% propyl alcohol was used as the electrophoretic medium and the standard uncoated fused silica capillary(75 μm×64.5 cm, 56 cm of effective length) was used as the separation pathway. The separation voltage was 20 kV with a detection wavelength of 280 nm; the column temperature was maintained at 25 ℃, and the sample was injected at 5 kPa, 6 s. Results: The above-mentioned six flavonoids showed good linearity(r>0.9990) in the range of the tested concentration, and the average recoveries of the method were between 96.20%-103.6%. Conclusion: The method is simple, accurate and reproducible, and can be used for quality control and evaluation of Scutellariae Radix.
