目的: 建立HPLC多波长法测定刺五加颗粒中5个化合物(原儿茶酸、刺五加苷B、绿原酸、刺五加苷E和异嗪皮啶)的含量。 方法: 采用Agilent Zorbax SB-C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)-0.5%醋酸溶液(B)为流动相,梯度洗脱(0~10 min,10%A;10~20 min,10%A→13.5%A;20~70 min,13.5%A→15.5%A),流速0.6 mL·min-1,检测波长为270 nm(0~51 min,测定原儿茶酸、刺五加苷B、绿原酸和刺五加苷E)、345 nm(51~70 min,测定异嗪皮啶),柱温24 ℃。 结果: 原儿茶酸、刺五加苷B、绿原酸、刺五加苷E和异嗪皮啶浓度分别在0.935~187 μg·mL-1(r=1.0000)、1.52~303 μg·mL-1(r=1.0000)、2.74~547 μg·mL-1(r=1.0000)、3.62~723 μg·mL-1(r=1.0000)、0.510~102 μg·mL-1(r=1.0000)范围内呈良好线性关系;高、中、低浓度的平均回收率(n=3)分别为97.8%~102.6%、98.0%~103.5%、96.8%~104.8%,RSD分别为0.8%~4.0%、1.3%~4.0%、0.9%~4.0%。 结论: 该方法可用于刺五加颗粒的质量控制。

Objective: To establish a method for the simultaneous determination of five components (protocatechuic acid,eleutheroside B,chlorogenic acid,eleutheroside E and isofraxidin) in Ciwujia granules by HPLC with different UV wavelengths. Method: The analysis was performed on an Agilent Zorbax SB-C18column (4.6 mm×250 mm,5 μm) with a gradient elution (0-10 min,10%A;10-20 min,10%A→13.5%A;20-70 min,13.5%A→15.5%A) of acetonitrile (A)-0.05% acetic acid (B) at the flow-rate of 0.6 mL·min-1.The detection wavelength was 270 nm (0-51 min) for the determination of protocatechuic acid,eleutheroside B,chlorogenic acid and eleutheroside E,and 345 nm (51-70 min) for the determination of isofraxidin.The column temperature was at 24 ℃. Results: Protocatechuic acid,eleutheroside B,chlorogenic acid,eleutheroside E and isofraxidin had good linearities in the ranges of 0.935-187 μg·mL-1(r=1.0000),1.52-303 μg·mL-1(r=1.0000),2.74-547 μg·mL-1(r=1.0000),3.62-723 μg·mL-1(r=1.000 0),and 0.510-102 μg·mL-1(r=1.0000),respectively.The average recoveries of high,median,and low concentrations were 97.8%-102.6%,98.0%-103.5%,96.8%-104.8% and RSDs were 0.8%-4.0%,1.3%-4.0%,0.9%-4. 0%,respectively. Conclusion: It indicates the developed method can be used for the quality control of Ciwujia granules.
