目的: 建立郁舒颗粒HPLC-ESI-MS分析方法,鉴定其化学成分。 方法: 高效液相色谱采用Agilent ZORBAX SB-C18色谱柱(4.6 mm×250 mm,5 μm),以0.1%醋酸水溶液(A)-乙腈(B)为流动相,梯度洗脱,检测波长为203 nm;质谱使用ESI离子源,正、负离子分别检测。 结果: 通过质谱分析以及参照相关文献共鉴定出46个成分,其中木脂素类成分12个、黄酮苷类成分8个、单萜苷类成分7个、皂苷类成分7个、有机酸类成分4个、间苯三酚类成分4个、鞣质类成分2个、糖酯类成分2个。 结论: 建立的方法灵敏度高、分离度好,为郁舒颗粒物质基础研究提供了一种快速准确的分析方法。

Objective: To identify Yushu granules by using high performance liquid chromatography coupled with electrospray tandem mass spectrometry (HPLC-ESI-MS). Methods: The analysis conditions were optimized as follows:Agilent ZORBAX SB-C18 column (4.6 mm×250 mm,5 μm),mobile phase (A) 0.1% acetic acid phase (B) acetonitrile,gradient elution,detection wavelength at 203 nm.The analysis was performed in both negative and positive ion mode. Results: A total of 46 compounds were simultaneously identified by ESI/MS through comparison with reference standards and literature data,including 12 lignoids,8 flavonoid glycosides,7 monoterpene glycosides,4 organic acids,4 phloroglucinol derivatives,2 tannins and 2 sugur esters. Conclusion: The method is sensitive,and provides good and rapid qualitative characterization of the Yushu granules composition.
