目的:建立雷公藤根木质部与根皮的色谱指纹图谱。 方法:采用高效液相色谱法分析。色谱柱:Inertsil ODS-SP(5 μm,4.6 mm×250 mm);流动相:乙腈-水,二元梯度洗脱;检测波长:220 nm;流速:1.0 mL·min-1。测定10批药材根木质部、根皮的色谱图,建立标准指纹图谱。 结果:雷公藤根木质部和根皮HPLC指纹图谱上的峰数和峰形基本相同,根木质部、根皮指纹图谱共有峰均有19个,但二者指纹图谱3、4 号共有峰的峰位和峰形不同。 结论:该分析方法的稳定性、精密度、重现性较好,为雷公藤药材根木质部与根皮的鉴别及质量控制提供了依据。

Objective: To develop a reliable high performance liquid chromatographic(HPLC)method for the fingerprint chromatogram study of the root xylems and barks from Tripterygium wilfordii Hook.F.,and compare the difference between them. Methods: HPLC separation was performed on an Inertsil ODS-SP column(5 μm,4.6 mm×250 mm)and detected by ultraviolet absorbance at 220 nm.A mobile phase composed of acetonitrile and H2O was adopted at a flow rate of 1.0 mL·min-1 with gradient elution.Ten batches of sample were determined respectively. Results: The peak profile of the 19 components collected within 120 min made up the Tripterygium wilfordii Hook.F.with universal features.The No.3 and No.4 common peaks in the fingerprints of the root xylems and barks were different. Conclusion: The fingerprint chromatograms had a good precision,stability,and reproducibility.The method is suitable for differentiation of the root xylems and barks from Tripterygium wilfordii Hook.F.,the present study provided a new evidence for the criterion of preparation process.
