目的: 建立HPLC同时测定无梗五加 果实中金丝桃苷和槲皮素-4'-O-β-D-半乳糖苷含量的方法。 方法: 采用TopsilTM(拓谱)C18(5 μm,4.6 mm×250 mm)色谱柱,流动相为乙腈-0.2%乙酸水溶液(体积比为17.5:82.5),流速1.0 mL·min-1,检测波长360 nm。 结果: 金丝桃苷的线性范围为0.036~0.356 μg(r=0.9998),平均回收率为98.5%(RSD=2.1%),最低检测限为0.053 μg·mL-1,最低定量限为0.178 μg·mL-1;槲皮素-4'-O-β-D-半乳糖苷的线性范围为0.846~8.456 μg(r=0.9995),平均回收率为99.5%(RSD=1.9%),最低检测限为0.126 μg·mL-1,最低定量限为0.423 μg·mL-1。供试品溶液48 h内稳定,金丝桃苷和槲皮素-4'-O-β-D-半乳糖苷峰面积的RSD分别为2.4%和2.6%。 结论: 方法可用于无梗五加果实的质量控制。

Objective: To develop an HPLC method for the simultaneous determination of hyperin and quercetin-4'-O-β-D-galactoside in the fruits of Acanthopanax sessiliflorus(Rupr.et Maxim.)Seem.. Methods: A TopsilTM C18 column(5 μm,4.6 mm×250 mm)was used with the mobile phase of acetonitrile-0.2% acetic acid(17.5:82.5)at a flow rate of 1.0 mL·min-1,and the UV detection was set at 360 nm. Results: The linear range of hyperin was 0.036-0.356 μg(r=0.9998),the average recovery was 98.5%(RSD=2.1%),LOD was 0.053 μg·mL-1,and LOQ was 0.178 μg·mL-1; The linear range of quercetin-4'-O-β-D-galactoside was 0.846-8.456 μg(r=0.9995),the average recovery was 99.5%(RSD=1.9%),LOD was 0.126μg·mL-1,and LOQ was 0.423 μg·mL-1.The sample solution was stable for at least 48 h,RSDs of hyperin and quercetin-4'-O-β-D-galactoside were 2.4% and 2.6% respectively.
