HPLC同时测定苍耳子中6个化学成分的含量

目的: 用HPLC同时测定苍耳子中新绿原酸、绿原酸、隐绿原酸、噻嗪双酮苷、1,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸6个化学成分的含量。方法: 采用Eclipse XDB-C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇-0.1%磷酸为流动相进行梯度洗脱,流速1 mL·min-1,检测波长254 nm,柱温35 ℃。结果: 新绿原酸、绿原酸、隐绿原酸、噻嗪双酮苷、1,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸色谱峰分离良好。进样量分别在0.012~1.2、0.028~2.8、0.012~1.2、0.014~1.4、0.024~2.4、0.011~1.1 μg范围内线性关系良好。平均回收率(n=6)分别为102.3%、96.4%、99.1%、103.1%、103.0%、103.4%,RSD分别为1.8%、1.1%、1.8%、1.5%、2.0%、1.9%。结论: 建立的苍耳子HPLC含量测定方法可用于苍耳子的质量控制。

Objective: To establish a method for simultaneous determination of 6 constituents (neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, xanthiside, 1, 5-dicaffeoylquinic acid, and 4, 5-dicaffeoylquinic acid) in Fructus Xanthii. Methods: The assay was conducted on a Eclipse XDB-C18 column (250 mm×4.6 mm, 5 μm) with methanol-0.1% H3PO4 as mobile phase. The flow rate was 1.0 mL·min-1, the detection wavelength was 254 nm and the column temperature was 35 ℃. Results: The correlation coefficient between concentration and chromatographic peak area of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, xanthiside, 1, 5-dicaffeoylquinic acid, and 4, 5-dicaffeoylquinic acid was over 0.9999 in the range of 0.012-1.2, 0.028-2.8, 0.012-1.2, 0.014-1.4, 0.024-2.4, 0.011-1.1 μg, respectively. The average recoveries (n=6) of 6 constituents were 102.3%, 96.4%, 99.1%, 103.1%, 103.0%, and 103.4%, respectively; RSDs were 1.8%, 1.1%, 1.8%, 1.5%, 2.0%, and 1.9%, respectively. Conclusion: The established method can be used for the quality control of Fructus Xanthii.
