星点设计-效应面法优化壮药千斤拔提取工艺

目的: 采用星点设计-效应面法优化千斤拔的回流提取工艺。方法: 以乙醇浓度、提取时间和溶媒比为自变量,以染料木苷、染料木素和浸膏等的收率为因变量,通过对自变量的总评归一值进行多元线性回归和二项式拟合,采用效应面优化最佳的工艺条件。 结果: 各指标的二项式拟合方程均优于多元线性回归方程,确定回流提取工艺条件为乙醇浓度(X1)为60%~80%,溶媒比(X2)为24~26倍,提取时间(X3)为180~240 min,提取次数是2次。工艺验证的总评归一预测值与实际值的偏差为-7.2%,二项式拟合的复相关系数R=0.9343。 结论: 采用星点设计-效应面法优选方法优化的染料木苷和染料木素提取工艺,方法简便、预测性良好。

Objective: To optimize the extracting process for the roots of Moghania philippinensis(Merr.et Rolfe) Li.by central composite design and response surface method. Methods: Ethanol concentration,extraction time and ratio to solvent were selected as the independent variables;genistin,genistein content and extract yield were set as the dependent variables.Linear or nonlinear mathematic models were used to estimate the relationship between independent and dependent variables.The response surface method was used to optimize the extraction process of the roots of Moghania philippinensis. Results: The multiple nonlinear equations of each indicator were better than the linear equation.The optimal conditions for the extraction process were determined as follows:60%-80% aqueous ethanol (X1),180-240 min (X3) for one extraction,ratio to solvent (X2) 24:1 to 26:1,and extraction for 2 times.The deviation between observed and predicted values was-7.2%.The multiple correlation coefficient of binomial fitting of the complex model was as high as 0.9343. Conclusion: The central composite design and response surface method is convenient and highly predictive for optimizing the extraction process of the roots of Moghania philippinensis.
