目的: 以常春藤皂苷元和齐墩果酸为指标成分,建立HPLC内标法研究七叶莲药材质量评价方法。 方法: 采用Phenomenex C18柱(250 mm×4.6 mm,5 μm),以乙腈-甲醇-0.5%醋酸铵(55: 25: 20)为流动相,流速0.8 mL·min-1,以苯丙酸诺龙为内标,在210 nm检测水解后的2个皂苷元。 结果: 常春藤皂苷元和齐墩果酸浓度均在0.025~0.75 mg·mL-1(r=0.9993)范围内线性关系良好,平均加样回收率(n=9)分别为99.3%和98.1%。 结论: 10批不同来源七叶莲药材中常春藤皂苷元和齐墩果酸的含量测定结果表明,所建方法准确可靠,专属性好,可作为七叶莲药材的质量控制方法。

Objective: To develop an HPLC method for determination of hederagenin and oleanolic acid in Schefflera arboricola using nandrolone phenylpropionate as internal standard. Method: The chromatographic separation was achieved on a Phenomenex C18 column(250 mm×4.6 mm, 5 μm) with the mobile phase consisting of acetonitrile-methanol-0.5% ammonium acetate(55: 25: 20) at a flow rate of 0.8 mL·min-1. Hederagenin and oleanolic acid were determined after acid hydrolysis at the detection wavelength of 210 nm. Results: The linear ranges for both triterpenoid sapogenins were 0.025-0.75 mg·mL-1(r=0.9993). The average recoveries(n=9) of hederagenin and oleanolic acid were 99.3% and 98.1%, respectively. Conclusion: The established method was applied for the assay of hederagenin and oleanolic acid in 10 batches of Schefflera arboricola samples, which suggests the method is sensitive, reliable and applicable for the quality control.
