目的: 建立同时测定白及多糖分子量和含量的高效分子排阻色谱分析方法。 方法: 采用TSK-gel G4000 PWXL色谱柱(10 μm,7.8 mm×300 mm),流动相为纯水,流速0.6 mL·min-1,柱温30 ℃,示差折光检测器检测。 结果: 白及多糖重均分子量的线性范围为24.17~178.0 k(R2=0.9943),含量测定线性范围为8.22~20.55·mg·mL-1 (R2=0.9992),分子量、含量精密度试验RSD分别为0.11%和0.96%(n=6),重复性RSD为0.28%和1.1%(n=5)。 结论: 该方法简便快速,结果准确,重复性好,可作为白及多糖分子量和含量测定的有效方法。

Objective: To develop a high performance size exclusion chromatography method for the determination of molecular weight and content of polysaccharide from Bletilla striata. Methods: The TSK-gel G4000 PWXL (10 μm,7.8 mm× 300 mm) column was used at 30 ℃,with water as the mobile phase at the flow rate of 0.6 mL·min-1.Refractive index detector was applied. Results: The average molecular weight of Bletilla striata polysaccharide ranged from 24.17 k to 178.0 k (R2=0.9943).The content of Bletilla striata polysaccharide ranged from 8.22 to 20.55 mg·mL-1 (R2=0.9992).Relative standard deviations (RSDs) of measurement precision of molecular weight and content were 0.11% and 0.96% (n=6),respectively.RSDs of the repeatability were 0.28%,and 1.14% (n=5),respectively. Conclusion: The method is simple,rapid and effective for the measurement of molecular weight and content of Bletilla striata polysaccharides.
