目的: 建立一种用新型键合纤维素手性固定相拆分贝凡洛尔对映体的高效液相色谱方法。 方法: 使用Chiralpak IB柱(250 mm×4.6 mm,5 μm),流动相为乙腈-20 mmol·L-1 磷酸二氢钾溶液(25: 75,pH 4.5),流速0.8 mL·min-1,检测波长259 nm,柱温25℃,对盐酸贝凡洛尔对映体进行拆分,同时也在正相模式和极性有机模式下,对其进行拆分。 结果: 在3种模式下,盐酸贝凡洛尔在正相模式和反相模式下能够达到基线分离,在极性有机模式下能部分分离。 结论: 该实验方法操作简便,准确度高,重复性好。

Objective: To develop a liquid chromatographic method for the chiral separating enantiomers of bevantolol hydrochloride. Method: Bevantolol hydrochloride enantiomers were separated by a Chiralpak IB(250 mm×4.6 mm,5 μm) column at 25℃ and detected at 259 nm with the mobile phase of acetonitrile-20 mmol·L-1 potassium dihydrogen phosphate solution(25: 75,pH 4.5) at a flow rate of 0.8 mL·min-1,and the enantiomers were also separated in NP-mode and polar organic mode. Results: In RP-mode and NP-mode,enantiomers of bevantolol hydrochloride could reach baseline separation,and in polar organic mode,they could achieve partial separation. Conclusion: The method is simple and reproducible for separation of bevantolol enantiomers on Chiralpak IB column in the reversed-phase mode.
