目的: 建立反相高效液相色谱法快速测定丹参中丹酚酸B、二氢丹参酮Ⅰ、隐丹参酮、丹参酮Ⅰ和丹参酮ⅡA 含量。 方法: 采用Kinetex核-壳技术色谱柱(100 mm×4.6 mm,2.6 μm),以乙腈-0.1%三氟乙酸水溶液为流动相,梯度洗脱,柱温30℃,检测波长为280 nm(丹酚酸B)和254 nm(丹参酮类)。 结果: 丹酚酸B、二氢丹参酮Ⅰ、隐丹参酮、丹参酮Ⅰ和丹参酮ⅡA 5个成分浓度分别在3.40~850 μg·mL-1 (r=0.9998),0.96~240 μg·mL-1 (r=0.9999),2.04~255 μg·mL-1 (r=0.9995),0.27~68 μg·mL-1 (r=0.9998),1.12~210 μg·mL-1 (r=0.9997)范围内与峰面积呈现良好的线性;平均回收率(n=3)在97.32%~104.9%之间,RSD≤4.7%。 结论: 该方法快速、简便、灵敏,可用于丹参药材的质量控制。

Objective: To establish a RP-HPLC method for the fast simultaneous determination of salvianolic acid B,dihydrotanshinone Ⅰ,cryptotanshinone,tanshinone Ⅰ,and tanshinone ⅡA in Radix et Rhizoma Salviae Miltiorrhizae samples. Methods: These five constituents were analyzed on a Kinetex core-shell technology high performance chromatography column(100 mm×4.6 mm,2.6 μm) by a gradient elution using 0.1% trifluoroacetic acid aqueous solution and acetonitrile as the mobile phase at 30℃ column temperature;the detection wavelength was set at 280 nm for salvianolic acid B and 254 nm for tanshinones. Results: The results showed good linearities in the range of 3.40-850 μg·mL-1 (r=0.9998) for salvianolic acid B,0.96-240 μg·mL-1 (r=0.9999) for dihydrotanshinone Ⅰ,2.04-255 μg·mL-1 (r=0.9995) for cryptotanshinone,0.27-68 μg·mL-1 (r=0.9998) for tanshinone Ⅰ,and 1.12-210 μg·mL-1 (r=0.9997) for tanshinone ⅡA;the average recoveries of the five constituents ranged between 97.32% and 104.9% (RSD≤4.7%). Conclusion: The method is rapid,simple and sensitive which can be used for the quality control of Radix et Rhizoma Salviae Miltiorrhizae.
