HPLC和UPLC法测定活血止痛制剂中阿魏酸

目的: 建立活血止痛散和活血止痛胶囊中阿魏酸含量的HPLC和UPLC测定方法。 方法: 采用Agilent TC-C₁₈和AQUITY UPLC BEH C₁₈色谱柱,以甲醇-0.1%醋酸(30: 70)为流动相,流速分别为1.0 mL·min和0.2 mL·min,检测波长321 nm,柱温30℃。 结果: 阿魏酸在0.2~200 μg·mL之间线性关系良好,HPLC和UPLC检出限分别为0.54 ng和0.13 ng,平均回收率约为100%,RSD均小于3%。配对t检验结果显示HPLC和UPLC测定值无显著性差异。 结论: 方法简便、快速、灵敏、准确,可更好地控制活血止痛制剂的质量。

Objective: To establish HPLC and UPLC methods for determination of ferulic acid in Huoxue Zhitong powders and Huoxue Zhitong capsules. Methods: Separations were performed on an Agilent TC-C₁₈ column and an AQUITY UPLC BEH C₁₈ column,respectively.Mobile phase consisted of methanol-water containing 0.1% acetic acid(30: 70)and the flow rate was 1.0 mL·min^-1 and 0.2 mL·min^-1 separately.The detection wavelength was 321 nm and the column temperature was 30℃. Results: Calibration curve for ferulic acid was linear in the range of 0.2-200 μg·mL^-1,limits of detection were 0.54 ng and 0.13 ng for HPLC and UPLC methods.The average recoveries were around 100% with RSD below 3%.Paired t-test showed that the difference between the results determined with the two methods was not significant. Conclusion: The proposed method was simple,fast,sensitive,accurate and applicable for better control of Huoxue Zhitong preparations’ quality.
