HPLC法测定脂肪乳注射液及卵磷脂中溶血磷脂的含量

目的: 建立高效液相色谱法测定脂肪乳注射液及卵磷脂中溶血磷脂。 方法: 采用Alltima Silica色谱柱,以甲醇-冰醋酸(100∶0.2,三乙胺调节pH至6.0)为流动相,使用蒸发光散射检测器,漂移管温度:70 ℃,气体:空气,流量:1.8 L·min^-1。 结果: 在选定的条件下,溶血磷脂与样品中其他组分分离良好。溶血磷脂的检测限为0.1 μg;溶血磷脂在0.04~0.20 mg·mL^-1范围内,峰面积的对数与浓度的对数呈良好的线性关系;平均回收率为103.0%,RSD为1.8%。 结论: 本法简便、专属、准确,可用于脂肪乳注射液及卵磷脂中溶血磷脂的含量测定。

Objective: To develop an HPLC method for determination of lysophosphatidyl choline in fat emulsion injection and lecithin. Method: The HPLC method was performed on an Alltima Silica column with methanol-acetic acid(100∶0.2,adjusted pH 6.0 with triethylamine) as mobile phase,and the detector was ELSD,of which the drift tube temperature was maintained at 70 ℃ and the flow rate of carrier gas was kept at 1.8 L·min^-1. Result: Under the described condition,lysophosphatidyl choline was completely separated from other components in fat emulsion injection and lecithin.The LOD of lysophosphatidyl choline was 0.1 μg.The linear range of lysophosphatidyl choline was 0.04-0.20 mg·mL^-1,and the average recovery was 103.0%,RSD=1.8%(n=9). Conclusion: The method is simple,specific and accurate,and can be used for determination of lysophosphatidyl choline in fat emulsion injection and lecithin.
