目的: 建立同时测定双黄连冻干粉中11个成分(绿原酸、咖啡酸、芦丁、金丝桃苷、连翘酯苷、野黄芩苷、黄芩苷、连翘苷、木犀草素、黄芩素和汉黄芩素)含量的高效液相色谱法。 方法: 采用Agilent Zorbax SB-C18(250 mm×4.6 mm,5 μm)色谱柱,流动相为乙腈-0.1%甲酸水溶液,线性梯度洗脱,流速为1.0 mL·min-1;金丝桃苷、黄芩苷、连翘苷、黄芩素及汉黄芩素的检测波长为278 nm,绿原酸、咖啡酸、芦丁、连翘酯苷、野黄芩苷及木犀草素的检测波长为320 nm。 结果: 11个成分在考察的线性范围内,进样量与峰面积之间线性关系良好(r>0.9995);回收率(n=9)均在96.3%~104.0%范围内,RSD小于2.3%。 结论: 方法操作简单、准确,专属性强,适用于双黄连冻干粉的质量控制。

Objective: To establish an HPLC method for determination of eleven compounds(chlorogenic acid, caffeic acid, rutin, hyperoside, forsythiaside, scutellarin, baicalin, forsythin, luteoloside, baicalein and wogonin) in Shuanghuanglian freeze-dried powder. Methods: Agilent Zorbax SB-C18 column(250 mm×4.6 mm, 5 μm) was adopted. The mobile phase was composed of acetonitrile and 0.1% formic acid water-solution in gradient elution mode. The flow rate was 1.0 mL·min-1. The detection wavelength for hyperoside, baicalin, forsythin, baicalein and wogonin was set at 278 nm, while that for chlorogenic acid, caffeic acid, rutin, forsythiaside, scutellarin and luteoloside was 320 nm. Results: The calibration curves were linear in certain ranges for eleven compounds(r>0.9995), respectively. All recoveries(n=9) were within the range of 96.3%-104.0% with RSD values less than 2.3%. Conclusion: The method is simple and accurate with high specificity. It can be used for the quality control of Shuanghuanglian freeze-dried powder.
