目的: 建立高速逆流色谱分离纯化紫苏叶中迷迭香酸的方法。 方法: 采用高速逆流色谱分离纯化紫苏叶乙酸乙酯萃取部分中迷迭香酸,以石油醚-乙酸乙酯-甲醇-0.5%醋酸水溶液(2∶ 5∶ 2∶ 5)为溶剂体系,上相为固定相,下相为流动相,流速2.0 mL·min-1,主机转速800 r·min-1,温度25 ℃。 结果: 每0.20 g紫苏叶乙酸乙酯萃取物经1次纯化,纯度为98.6%的迷迭香酸的得率约13%。 结论: 高速逆流色谱可高效分离纯化紫苏叶中的迷迭香酸。

Objective: To establish a method for separation and purification of rosmarinic acid from Perilla frutescens(L.)Britt.var.acuta(Thunb.)Kudo form.discolor(Makino)Kudo(abbr:PFAD)by high-speed counter-current chromatography(HSCCC ). Methods: The compound was separated and purified by HSCCC from the ethyl acetate part of the crude extract of PFAD,using petroleum ether-ethyl acetate-methanol-0.5% acetic acid water-solution(2∶ 5∶ 2∶ 5)as the two-phase solvent system.The upper phase was chosen as the stationary phase,and the lower phase was chosen as the mobile phase.The mobile phase was operated at a flow rate of 2.0 mL·min-1,while the apparatus rotated at 800 r·min-1 and the temperature was set at 25 ℃. Results: The yield of rosmarinic acid with 98.6% purity was 13% from each 0.20 g ethyl acetate part of the crude extract of PFAD by once separation. Conclusion: HSCCC is a efficient method for separating,and purificating rosmarinic acid from PFAD.
