目的: 建立超高效液相色谱法检测小叶黑柴胡的根中柴胡皂苷a、d含量。 方法: 采用Waters Acquity UPLC系统,使用ACQUITY UPLC BEHTM C18(50 mm×2.1 mm,1.7 μm)色谱柱,流动相为乙腈-水,梯度洗脱,流速0.6 mL·min-1,检测波长203 nm,柱温30 ℃。 结果: 柴胡皂苷a线性范围为0.059~1.180 μg(r=0.9998),平均回收率为96.7%(RSD=1.3%,n=6);柴胡皂苷d线性范围为0.112~2.230 μg(r=0.9997),平均回收率为98.7%(RSD=1.5%,n=6)。 结论: 该方法操作简便,结果准确,可用于小叶黑柴胡的质量控制。

Objective: To establish an UPLC method with Photo Diode Array for simultaneous determination of saikosaponin a,saikosaponin d in roots of Bupleurum smithii Wolff var. parvifoliaum Shan et Y. Li. Methods: The analyses were performed on a Waters Acquity UPLC system, an ACQUITY UPLC BEHTM C18(50 mm×2.1 mm,1.7 μm) column was adopted,the mobile phase consisted of acetonitrile-water with gradient elution at the flow rate of 0.6 mL·min-1,the detection wavelength was set at 203 nm,and the column temperature was 30 ℃. Results: The linear ranges for saikosaponin a,saikosaponin d were 0.059-1.180 μg(r=0.9998) and 0.112-2.230 μg(r=0.9997),with their average recoveries(n=6) of 96.7%(RSD=1.3%) and 98.7%(RSD=1.5%),respectively. Conclusion: The method is simple,rapid and accurate,and can be used for the quality control of Bupleurum smithii Wolff var. parvifoliaum Shan et Y. Li.
