目的: 建立测定扛板归不同部位中齐墩果酸和熊果酸含量的高效液相色谱-二极管阵列检测器方法。方法: 色谱柱为Kromasil C18柱(4.6 mm×250 mm,5 μm),流动相为0.5%磷酸水溶液-甲醇(10∶[KG-*2]90),流速0.8 mL·min-1,检测波长210 nm,柱温30 ℃。结果: 齐墩果酸在0.228~2.850 μg时,与峰面积呈良好的线性关系(r=0.9998),平均回收率为 97.4%,RSD为1.5%(n=6);熊果酸进样量在0.544~6.800 μg时,与峰面积呈良好的线性关系(r=0.9998),平均回收率为 96.9%,RSD为1.2%(n=6)。结论: 方法准确,操作简便,数据可靠,可用于扛板归中齐墩果酸和熊果酸的含量测定。

Objective: To establish a HPLC-DAD method for the determination of oleanolic acid and ursolic acid in the different parts of Polygonum perfoliatum L. Methods: Kromasil C18 column(4.6 mm×250 mm,5 μm)was adopted.The mobile phase consisted of methanol-0.5% phosphoric acid aqueous solution(10∶90)at a flow rate of 0.8 mL·min-1,and the column temperature was set at 30 ℃.The detection wavelength was 210 nm.Results: Oleanolic acid showed good relationship at the range of 0.228-2.850 μg(r=0.9998)and the recovery was 97.4% with 1.5%(n=6)of RSD.Ursolic acid showed good relationship at the range of 0.544-6.800 μg(r=0.9998)and the recovery was 96.9% with 1.2%(n=6)of RSD.Conclusion: The proposed method is accurate,simple,and promises to be applicable for the determination of oleanolic acid and ursolic acid in the different parts of P.perfoliatum L.
