目的:建立测定Beagle犬血浆中四环素的RP-HPLC分析方法。方法:Beagle犬血浆样品经001 mol·L-1草酸酸化、乙腈沉淀蛋白处理后,采用Diamonsil C18柱(200 mm × 4.6 mm,5 μm)分离,流动相为乙腈-0.01 mol·L-1草酸溶液(15∶85),流速为0.8 mL·min-1,检测波长为350 nm,内标为土霉素。结果:测定血浆中四环素的线性范围为0.2 ~ 10 μg·mL-1;日内与日间精密度均小于14.5%,准确度(RE)在 ±5.3 %以内。结论:本方法简便、灵敏、准确,适用于Beagle犬血浆中四环素的分析测定。

Objective:To develop an RP-HPLC method for the determination of tetracycline in Beagle dog plasma.Methods:After acidification with oxalic acid,plasma samples were precipitated with acetonitrile.Chromatographic separation was carried out on a Diamonsil C18 column(200 mm × 4.6 mm,5 μm)using a mobile phase of acetonitrile-0.01 mol·L-1oxalic acid(15∶85).The flow rate was 0.8 mL·min-1 and the detection wavelength was 350 nm.Oxytetracycline was used as the internal standard.Results:The calibration curve for tetracycline was linear over the concentration range of 0.2-10 μg·mL-1.The intra- and inter-day precision was less than 14.5% and the accuracy was within ± 5.3%.Conclusion:The method is simple,sensitive,accurate and suitable for the preclinical pharmacokinetic studies of tetracycline in dog plasma.
