目的: 建立舒肝平胃丸中厚朴酚在大鼠体内血药浓度的LC-MS/MS测定方法,并研究其在大鼠体内的药动学。 方法: 以1,8-二羟基蒽醌为内标,血浆样品经甲醇沉淀蛋白。以Agilent ZOBAX SB-C18 柱(2.1 mm×50 mm,3.5 μm)进行分离;采用电喷雾离子源(ESI源)以正离子方式检测;扫描方式:多反应离子监测(MRM)方式。 结果: 厚朴酚血药浓度在0.0166~1.66 μg·mL-1范围内线性关系良好(r=0.9996);平均提取回收率在83.3%~86.1%之间,日内、日间RSD均≤7.2%。 结论: 本方法快速、简便、准确、灵敏,适用于舒肝平胃丸中厚朴酚的药动学研究。

Objective: To establish an LC-MS/MS method for the determination of Magnolol concentration of Shugan Pingwei Pills in plasma and to study its pharmacokinetics in rat. Methods: 1,8-2-hydroxy anthraquinone as the internal standard,plasma proteins were precipitated with methanol.Agilent ZOBAX SB-C18 column (2.1 mm×50 mm,3.5 μm) was used to separate.Ion source:ESI ion source (ESI source);The LC-MS system was operated using an electrospray ionization probe in the positive ion mode;Scan mode:multiple reaction ion monitoring (MRM) mode. Result: The liner range of magnolol was 0.0166-1.66 g·mL-1 (r=0.9996),average extracted recovery at 83.3%-86.1%,and RSD of inter-day and intra-day at no more than 7.2%. Conclusion: The method developed in the study was proved to be rapid,simple,accurate and sensitive,and suitable for the pharmacokinetic study of Magnolol of Shugan Pingwei Pills in rats.
