目的: 建立RP-HPLC-UV法测定5种发酵虫草制剂百令胶囊、宁心宝胶囊、心肝宝胶囊、至灵胶囊及金水宝胶囊中麦角甾醇的含量。 方法: 色谱柱为Agilent SB TC C18(250 mm×4.6 mm,5 μm),检测波长283 nm,流动相为甲醇-水(98∶ 2),流速1.0 mL·min-1。 结果: 麦角甾醇进样量在0.42~6.29 μg范围内呈良好的线性关系(r=0.9999)。平均加样回收率(n=3)分别为98.1%,100.7%,99.6%;RSD分别为2.3%,1.6%,1.6%。18批发酵虫草制剂中麦角甾醇含量,百令胶囊为6.06~6.10 mg·g-1,宁心宝胶囊为2.51 ~3.93 mg·g-1,心肝宝胶囊为3.47~4.42 mg·g-1,至灵胶囊为2.48~4.61 mg·g-1,金水宝胶囊为3.45~3.87 mg·g-1。 结论: 本文方法操作简便、灵敏,具有良好的重复性和稳定性,可用于发酵虫草制剂中麦角甾醇的检测。

Objective: To develop an RP-HPLC-UV method for the determination of contents of ergosterol in the preparations of fermental Cordyceps,such as Bailing capsule,Ningxinbao capsule,Xinganbao capsule,Zhiling capsule and Jinshuibao capsule. Method: The analysis was carried out on an Agilent SB TC C18(250 mm×4.6 mm,5 μm) column eluted with the mobile phase consisting of methanol-water (98∶ 2).The flow rate was 1.0 mL·min-1 and the detection wavelength was set at 283 nm. Results: The linear range of ergosterol was 0.42-6.29 μg (r=0.9999).The average recoveries (n=3) were 98.1%,100.7%,99.6% and RSDs were 2.3%,1.6%,1.6% respectively.The contents of ergosterol in preparations of fermental Cordyceps were 6.06-6.10 mg·g-1 (Bailing capsule),2.51-3.93 mg·g-1 (Ningxinbao capsules),3.47-4.42 mg·g-1 (Xinganbao capsules),2.48-4.61 mg·g-1 (Zhiling capsules) and 3.45-3.87 mg·g-1 (Jinshuibao capsules). Conclusion: The method is effective,sensitive and accurate with high repeatability and stability,which is helpful for the determination of ergosterol in the preparations of fermental Cordyceps.
