目的: HPLC法定量分析不同产地、不同生长年限的刺五加药材中异嗪皮啶。 方法: 采用高效液相色谱法,色谱柱:Kromasil C18分析柱(200 mm×4.6 mm,5 μm);流动相:乙腈-0.1%磷酸(20∶ 80);流速:1 mL·min-1;检测波长:343 nm;柱温:30 ℃;进样量20 μL。 结果: 异嗪皮啶进样量在0.0171~0.3024 μg范围内呈现良好的线性关系,平均回收率为99.4%,RSD为2.9%(n=9)。铁西产刺五加根中异嗪皮啶含量2年生最高(110.15 μg·g-1),茎中1年生最高;么河产不同生长年限刺五加中含量差别不大;根中异嗪皮啶的含量明显高于茎;铁西产刺五加中异嗪皮啶含量比么河产高。 结论: 本法操作简便、准确,专属性强,适用于刺五加中异嗪皮啶含量测定,该研究可为刺五加最佳产地、最适宜采收年限及最佳药用部位的确定提供参考。

Objective: To establish an HPLC method for determination of isofraxidin in Acanthopanax senticosus (Rupr.et Maxim.)Harms of different sources and growth time limits. Methods: The Kromasil C18 reversed-phase column (200 mm×4.6 mm,5 μm) was adopted with the mobile phase of acetonitrile-0.1% phosphoric acid(20∶ 80),at the flow rate of 1 mL·min,-1the detection wavelength 343 nm,the column temperature was 30 ℃ and the injection volume was 20 μL. Results: Isofraxidin had a good linearity in the range of 0.0171-0.3024 μg.The average recovery of isofraxidin was 99.4%,RSD=2.9%(n=9);The two-year Acanthopanax senticosus of Tiexi had the highest isofraxidin content in root(110.15 μg·g-1),the one-year Acanthopanax senticosus of Tiexi had the highest isofraxidin content in stem;The content of isofraxidin in Acanthopanax senticosus from Mehe of different growth time had little difference,and the content in root was significantly higher than that in stem;The content of isofraxidin of Acanthopanax senticosus from Tiexi was higher than that from Mehe. Conclusion: This developed method is simple,accurate and high specificity,suitable for quantitative determination of isofraxidin in Acanthopanax senticosus.The study provides the reference for the best origin,the most rational harvest time and medicinal parts of Acanthopanax senticosus.
