目的: 用高效凝胶渗透色谱 (SEC)和高效凝胶渗透色谱及激光光散射联用(SEC-MALLS)法测定玻璃酸钠的相对和绝对分子质量,并对2种分析方法进行比较。 方法: 色谱柱为SB806HQ,流动相为0.2 moL·L-1的氯化钠溶液(含0.02%叠氮化钠),流速为0.5 mL·min-1,柱温为35 ℃,进样量100 μL, SEC法用示差折光检测器,SEC-MALLS法用DNDC仪和激光光散射仪联用检测。 结果: 不同样品SEC法和SEC-MALLS法测得的分子质量结果接近,并且测定结果在一定范围内与样品浓度有关,浓度对相对分子质量的影响更大。 结论: SEC和SEC-MALLS法均可用于玻璃酸钠分子质量测定,浓度选择0.1 mg·mL-1为宜。

Objective: To determine the relative and absolute molecular weight of sodium hyaluronate(SH)by SEC and SEC-MALLS methods,and then compare the two analysis methods. Methods: SB806HQ column was used with mobile phases of 0.2 moL·L -1 sodium chloride(containing 0.2%sodium azide),at 35 ℃ with a flow rate 0.5 mL·min-1 by RI(used by SEC method),DNDC with MALLS detectors(used by SEC-MALLS method),the injection was 100 μL. Results: The Mw of SEC method(standards from Longzhida company) and SEC-MALLS method was similar,while the results were both concerned with the concentration of the samples in certain limit. Conclusion: The SEC method and SEC-MALLS method could both be used to determine the molecular weight of SH,and the suitable concentration were 0.1 mg·mL-1.
