目的: 建立脂肪乳注射液及卵磷脂中溶血磷脂酰胆碱(LPC)的薄层色谱检查方法。 方法: 样品用正庚烷-异丙醇(1∶2)溶解后直接点于硅胶G薄层板上;展开剂为氯仿-无水乙醇-三乙胺-水(10∶ 13∶ 5∶ 3),显色剂为Dittmer-Lester试剂;薄层扫描条件:双波长锯齿扫描,测定波长680 nm,参比波长400 nm,线性参数SX=3,狭缝宽度0.4 mm×0.4 mm,灵敏度中等。 结果: 在上述条件下,样品中LPC斑点与其他磷脂斑点分离良好,LPC在0.4~10 μg范围内线性良好,r=0.9997,平均加样回收率为99.4%,RSD=2.1%。 结论: 本法简便、快速、成本低,可用于脂肪乳注射液及卵磷脂中LPC的常规检查。

Objective: To establish a TLC method for determination of lysophosphatidyl choline in fat emulsion injection and phospholipids. Method: Sample dissolved in n-heptane-isopropyl alcohol (1∶ 2) was used as test solution,and can be spotted on Silica gel G thin-layer plate directly;developing solvent was chloroform-ethanol-triethylamine-water (10∶ 13∶ 5∶ 3) and Dittmer-Lester reagent was used for coloration;TLC scanning conditions:dual wavelength sawtooth scanning,detection wavelength was 680 nm,reference wavelength was 400 nm,linear parameter was SX=3,slit width was 0.4 mm×0.4 mm,sensitivity was moderate. Results: Under these conditions,lysophosphatidyl choline spot in samples can be completely separated from other phospholipid spots,LPC in the range of 0.4-10 μg showed good linearity,r=0.9997,average recovery was 99.4%,RSD=2.1%. Conclusion: The method,which can be used for determination of lysophosphatidyl choline in fat emulsion injection and phospholipids,is simple,rapid and economical.
