目的: 建立同时测定板蓝根颗粒中4种核苷类成分含量的快速液相色谱方法。 方法: 色谱柱:Agilent Zorbox SB C18(4.6×50 mm,1.8 μm),流动相:水(A)-甲醇(B)线性梯度洗脱 ,流速:0.6 mL·min-1,柱温:30 ℃,检测波长:254 nm。 结果: 尿苷的线性范围为2.164~86.56 μg,r=0.9997;鸟苷的线性范围为2.49~99.6 μg,r=0.9999;表告依春的线性范围为3.696~147.84 μg,r=0.9999;腺苷的线性范围为2.476~99.040 μg,r=0.9999。尿苷、鸟苷、表告伊春及腺苷平均回收率分别为为95.9%,99.9%,101.0%,99.8%,平均RSD分别为0.8%,1.8%,1.3%,0.8%(n=6)。5家企业的的板蓝根颗粒中核苷类成分的含量存在差异。 结论: 该方法操作简单,重复性好,灵敏度高,结果准确可靠。

Objective: To establish a quantitative method for determining the contents of four nucleosides in Banlangen Keli by UPLC. Method: The column was Agilent Zorbax SB C18(4.6 mm×50 mm,1.8 μm),the mobile phase consisted of water(A) and methanol(B) with linear gradient elution(0-4 min,0-3%B;4-12 min,3-12%B;12-16 min,50-50%B;16-20 min,0-0%B),the flow rate was 0.6 mL·min-1,the column temperature was 30 ℃,Detection wavelength was 254 nm. Result: The linear range of uridine,guanosine,epigoitrin and adenosine were 2.164-86.56 μg (r=0.9997);2.49-99.6 μg(r=0.9999);3.696-147.84 μg(r=0.9999);2.476-99.040 μg(r=0.9999),.Their average recoveries were 95.9%,99.9%,101.0%,99.8% with RSD=0.8%,1.8%,1.3%,0.8%%,respectively(n=6)。.The contents of nucleosides were different in samples of five productive enterprises. Conclusion: The method is efficient,good reproducible,sensitive,and accurate.
