目的: 建立手性固定相拆分安倍生坦的高效液相色谱方法。 方法: 用多糖衍生化手性固定相,考察了流动相中异丙醇比例、柱温、流速对安倍生坦及其对映体手性拆分的影响。采用Chiralpak AD-H色谱柱(250 mm×4.6 mm,5 μm),以正己烷-异丙醇(17.5∶ 82.5)为流动相,流速为0.4 mL·min-1,检测波长263 nm,柱温25 ℃,拆分安倍生坦及其对映体。 结果: Chiralpak AD-H色谱柱能使安倍生坦与对映体达到基线分离,(S)-对映体先于(R)-对映体出峰。 结论: 本法操作简便,可用于安倍生坦及其对映体的分离和质量控制。

Objective: To develop a chiral stationary phase-HPLC method for the enantiomeric separation of racemic ambrisentan. Method: Effects of isopropanol modifier concentration,temperature and flow rate with polysaccharide-based chiral column as a stationary phase were investigated.The enantiomers of ambrisentan were separated on a Chiralpak AD-H column(250 mm×4.6 mm,5 μm) with a mobile phase consisted of n-hexane-isopropanol(17.5∶ 82.5) at a flow rate of 0.4 mL·min-1.The wavelength detector was set at 263 nm. Results: A baseline separation of ambrisentan enantiomers was achieved on the Chiralpak AD-H column.And the (S)-enantiomer eluted prior to (R)-one. Conclusion: It is recommended that this sample and selected method can be used for determination and quality control of ambrisentan and its enantiomer.
