中药羌活质量标准研究___

Study on the quality standard for Notopterygii Rhizoma et Radix

目的: 研究提高中药羌活的质量标准。 方法: 照2005年版中国药典方法检查中药羌活中总灰分和酸不溶性灰分,测定其挥发油含量;采用TLC法鉴别紫花前胡苷及RP-HPLC法测定羌活醇和异欧前胡素的总量。TLC条件 : 含3%醋酸钠的羧甲基纤维素钠溶液制备的硅胶G薄层板,氯仿-甲醇(8∶ 2)为展开剂,紫外光灯(365 nm)下检视;HPLC条件:色谱柱为Phenomenex Luna C18 (2)(4.6 mm×150 mm,5 μm),流动相为乙腈-水(44∶ 56),流速1.0 mL·min-1,检测波长310 nm。 结果: 中药羌活中总灰分和酸不溶性灰分分别小于8.0%和4.0%,挥发油平均含量为2.08%,紫花前胡苷薄层斑点清晰、特征明显。羌活醇和异欧前胡素的线性范围分别为9.97~99.68 μg·mL-1(r=0.9999)和1.17~11.74 μg·mL-1(r=0.9999),平均加样回收率(n=6)分别为97.3%(RSD=1.0%)和99.3%(RSD=1.3%)。 结论: 本法操作简便,重现性好,结果准确,可作为中药羌活的质量控制方法。

Objective: To improve the quality standard of Notopterygii Rhizoma et Radix. Methods: According to Chinese Pharmacopoeia,the total ash,acid-insoluble ash and volatile oil contents were determined.The TLC and HPLC methods were adopted to identify nodakenin and determine the total amount of notopterol and isoimperatorin,respectively.TLC conditions:Using silica gel G mixed with sodium carboxymethylcellulose containing 3% sodium acetate as the coating substance and chloroform-methanol(8∶ 2)as the mobile phase.HPLC conditions:Using Phenomenex Luna C18(2)(4.6 mm×150 mm,5 μm)column as analytic column and acetonitrile-water(44∶ 56)as the mobile phase,with a flow rate of 1.0 mL·min-1 and UV detection at 310 nm. Results :The total ash and acid-insoluble ash were not more than 8.0% and 4.0%,the volatile oil content was 2.08% on average.The TLC identification was highly specific and spot of nodakenin was clear.The linear ranges of notopterol and isoimperatorin were 9.97-99.68 μg·mL-1(r=0.9999)and 1.17-11.74 μg·mL-1(r=0.9999),the average recoveries(n=6)were 97.3%(RSD=1.0%)and 99.3%(RSD=1.3%). Conclusion: The method is simple with specificity and good reproducibility and can be used for quality control for Notopterygii Rhizoma et Radix.
